摘 要:为了提高微生物肥料生产菌株——巨大芽胞杆菌1013的发酵产芽孢数量,采用响应面法对其发酵培养基进行优化。首先采用单因子试验筛选出最佳碳源和氮源;然后设计基础培养基,采用Plackett-Burman方法对基础培养基中影响发酵产芽胞数各因素的效应进行评价,筛选出有显著效应的因素;最后通过响应面分析法优化主要因素的最佳浓度。结果表明,最佳碳源为糖蜜,最佳氮源豆饼粉和硝酸铵;基础培养基中对产芽孢数有显著效应的因素为糖蜜和豆饼粉,响应面优化后这2个因素的最佳含量为:糖蜜4.17%,豆饼粉1.53%。采用优化后的培养基进行摇瓶发酵,巨大芽胞杆菌的发酵产芽胞数为(89.34±3.87)×108cfu/mL,比基础培养基提高了3.35倍。The response surface methodology was used to optimize the fermentation medium of Bacillus megaterium 1013. In the single factor experiment, molasses, soybean powder and NH4NO3, were selected to be the optimal carbon source and nitrogen source, respectively. Then, molasses and soybean powder were found to be the important factors by Plackett-Burman design. The optimal concentration of two important factors was determined by response surface methodology. The results showed that, 4.17% molasses, 1.53% soybean powder. The spores yield of Bacillus megaterium 1013 could reach (89.34±3.87) × 108 cfu/mL by using the optimal medium, which was increased by 3.35 fold compared with the original medium.
