摘 要:目的观察人胎盘间充质干细胞(PMSCs)经低温冻存复苏前与复苏后的生物学特性。方法采用胶原酶和胰蛋白酶消化胎盘,分离获得PMSCs传代培养,取第3代细胞加入冷冻保护剂,利用程控降温仪冷冻,置于-196℃液氮中冻存6个月后复苏,比较冻存前和复苏后PMSCs的细胞形态、生长曲线、及成骨、成脂诱导分化培养,检验其是否仍具有多向分化潜能。结果 PMSCs经低温保存复苏后,细胞仍保持贴壁生长等外形特征,仍呈成纤维样形态生长,复苏后PMSCs生长曲线与冻存前相似,复苏后HUC-MSCs具有向成骨、成脂细胞诱导分化能力,细胞经茜素红染色、油红O染色呈阳性反应。结论冻存复苏后PMSCs的生物学特性仍保持稳定,保持了间充质干细胞的基本形态,仍具有向成骨细胞和脂肪细胞分化的潜能。Objective To investigate the effect of cryopreservation on biological features of human placenta--derived mesenchymal stem cells(PMSCs). Methods PMSCs were isolated by collagenase digestion and subcultured in vitro. The third passage PMSCs were frozen with a controlled rate freezer and thawed 6 months after they were stored in -196 ℃ liquid nitrogen. Their biological features were observed. Results After cryopreservation,PMSCs remained the characteristics of adherent growth. PMSCs grew in a fibroblastic morphology after thawing with a similar growth curve to that before cryopreservation. PMSCs had the osteogenic,adipogenic cells in ducedability to differentiate,the positive for alizarin red,oil red O staining. Conclusion After cryopreservation and thawing, PMSCs maintained their biological features and had the potential to differentiate into bone cells and fat cell.
