摘 要:以脱细胞猪真皮基质材料的边角料为原料,使用酸-酶结合法提取I型胶原,并用SDS-PAGE电泳、DSC、FI-IR、SEM、AFM对Ⅰ型胶原进行了结构表征。研究表明:组织学观察发现,脱细胞猪真皮基质材料中不存在细胞和细胞碎片,且胶原纤维得到充分的分离和松散,非纤维成分大部分被水解,有利于Ⅰ型胶原的提取。提取得到的I型胶原红外特征峰明显,分子质量大且分布窄,变性温度为69.4℃;SEM显示胶原呈现孔隙均匀的三维网状结构;AFM观察到胶原大部分排列紧密,相互编织缠绕,形成无规线团,小部分胶原直径较粗,且以线性分布,说明胶原已经发生自聚集现象。综合检测分析结果,可以认为所提取制备得到的确为具有天然三股螺旋结构的I型胶原。The histological changes of fresh pigskin after acellular were observed and analyzed by hematoxylin - eosin staining ( HE ) and iron hematoxylin staining. Using the offcut of porcine acellular dermal matrix as a raw material, collagen was extracted with the method of acid swelling - pepsin digestion. Then the obtained collagen was characterized by SDS - PAGE, Fq' - IR spec- tra, Differential Scanning Calorimeter ( DSC } , Atomic Force Microscope ( AFM } and Scanning study shows that: the cells in fresh pigskin are removed completely after the treatment process, Electron Microscopic ( SEM ). The the collagen fibers are loosed well, and most of the non - fiber component is hydrolyzed, which is conducive to the extraction of collagen. The extracted type I colla- gen has its characteristic peaks of FI - IR, and the molecular weight is of not only large, but also narrow distribution. The denat- uration temperature is 69.4~C ; the SEM displayed that the collagen remains three - dimensional mesh structure with uniform pore. We could also conclude that the collagen has closely arranged, mutually braided or wound to form a random coil. The diameter of the small portion of the collagen is coarse, and is in a linear distribution, this indicates that the self - aggregation of collagen oc- curs. As a consequence, the collagen sustains original triple helix structure.
