摘  要:目的探讨肿瘤坏死因子-α(TNF-α)对心脏β3-肾上腺素能受体(β3-AR)表达的影响及其在心衰治疗中的意义。方法培养生后2 d SD大鼠心肌细胞,取第4天的细胞加入含不同浓度TNF-α的培养液,检测不同时间作用下β3-AR mRNA及蛋白表达。结果心肌细胞受TNF-α刺激后,β3-AR mRNA及蛋白随着TNF-α作用时间的延长、作用浓度的增加,表达量增加,至作用浓度为1.6 ng/mL、作用时间为12 h达峰值。结论TNF-α作用下心肌β3-AR上调,且有时间及浓度依赖性,这可能是引起心衰的原因之一。Objective To investigate the effects of TNF-α on β3-AR expressions of cardiac myocyte in vitro and its significance on heart failure therapy.Methods The myocardial cells of the 2-day-old SD rat was cultured.In the fourth day,the cardiomyocytes were treated with different concentrations of TNF-alpha,and the presence of β3-AR both at the mRNA level and at the protein level with a specific monoclonal antibody was demonstrated.Results Below the concentration of 1.6 ng / mL and before the time of 12 hours,TNF-alpha can cause a time-dependent and concentration-dependent increase in β3-AR expression of cardiomyocytes.Conclusion Treatment of cardiomyocytes w ith TNFalpha causes a time-dependent and concentration-dependent increase in β3-AR expression of cardiomyocytes.This change may contribute to cardiac dysfunction.
