摘  要:目的探讨丹参酮ⅡA对大鼠心肌纤维化的保护作用。方法 SD大鼠60只采用腹主动脉缩窄术制备压力负荷增加心肌纤维化模型,术后存活中的24只大鼠随机分为3组,假手术组、模型组、丹参酮ⅡA组(20mg/(kg.d),每组各8只。术后4周造模成功并开始给药,疗程为4周,8周后检测心肌质量指数、心肌组织病理学、心肌羟脯氨酸及心肌组织转化生长因子-β1蛋白水平。结果模型组大鼠心肌质量指数、心肌羟脯氨酸及心肌组织中转化生长因子-β1蛋白水平明显高于假手术组与丹参酮ⅡA组(P<0.01)。结论丹参酮ⅡA可抑制心肌肥厚,减轻心肌纤维化,可能与下调心肌组织转化生长因子-β1表达有关。Objective To explore the protective effects of tanshinone(Tan) Ⅱ A on myocardial fibrosis.Methods Models of myocardial fibrosis were established in 60 SD rats induced by the abdominal aorta constriction preparation,and 24 rats of which were survived and randomly divided into sham operation group,model group and Tan Ⅱ A group(20 mg/(kg·d)),with 8 rats in each group.The models were successfully established 4 weeks after operation.The heart mass index,histopathological changes,myocardial hydroxyproline content and myocardial ransforming growth factor(TGF)-β1 expression were measured after four weeks of Tan Ⅱ A treatment.Results The heart mass index,myocardial hydroxyproline content and myocardial TGF-β1 level were obviously higher in model group than those in sham operation group and Tan Ⅱ A group(P0.01).Conclusion Tan Ⅱ A can inhibit cardiac hypertrophy and attenuate myocardial fibrosis,which may be associated with the downregulation of the expression of TGF-β1.
