摘 要:【目的】通过在培养基中添加不同浓度配比的植物激素,优化以18-599为受体的玉米遗传转化体系,为规模化的转基因育种服务。【方法】以N6/MS培养基为基本培养基,选用西南地区玉米骨干自交系18-599为受体材料,分别在分化、壮苗和生根培养基中添加KT、S-3307及ABT等植物生长调节剂,比较不同激素的处理对转基因幼苗的分化、壮苗和生根的影响,同时,还对KMnO4处理后的转基因幼苗的移栽成活率进行了研究。【结果】结果表明:在分化培养基中添加0.5mg/L KT可以有效地提高愈伤的分化率;在生根培养基中添加1.0mg/L ABT+1.0mg/LIBA可显著幼苗生根率,最高可达80%;在壮苗培养基中添加0.5mg/L S-3307可促进幼苗根系发育,矮化植株,使植株生长健壮;移栽时用KMnO4短时间处理能显著提高玉米转基因幼苗移栽成活率,最高可达到73.3%。【结论】在培养基中添加合适浓度的植物激素能提高转基因幼苗的分化率和生根率,并达到壮苗效果。在移栽时用低浓度的KMnO4短时间处理,提高了转基因玉米幼苗成活率,从而使以18-599为受体的玉米转基因再生体系得到了优化。[Objective] This study optimized the transgenic system based on the acceptor of maize inbred line 18-599 through adding different kinds of plant hormones at different concentration into the medium, and aimed to facilitate large-scale of transgenic breeding. [Method] In this study, maize inbred line 18-599, a key inbred line in southwest, was performed as the transgenic receptor and N6/MS medium was used as the base medium. Several mediums modified by adding plant hormones including KT, ABT and S-3307 were compared in terms of callus differentiation, seedling vigor and root generation:. Meanwhile, the survival rate of transgenic seedlings was improved when exposed to KMnO4 in short time. [Results] The results suggested that N6 medium containing 0.5 mg/L KT could improve the frequency of callus differentiation. N6 medium containingl. 0 mg/L ABT and 1.0 mg/L IBA significantly increased root regeneration up to 80%. Modified N6 medium containing 0.5 mg/L S-3307 could stimulate the growth of root development and make seedlings dwarf and vigor. The survival rate of the transgenic seedlings can be raised up to 74. 2% when subjected to short time treatment of KMnO4. [Conclusion] In this study, treatment including adding suitable plant hormone in the medium could enhance callus differentiation, promote root growth, and improve seedling vigor. Meanwhile, exposing the seedling to low concentration KMnO4 on transplanting facilitated the survival rate of the transgenic seedlings. These resuits showed that the genetic transformation system based on the acceptor of maize inbred line 18- 599 was further optimized in this study.
