摘  要:目的:研究5-氟尿嘧啶植入剂(5-Fuimplant5Ful)犬皮下植入的药代动力学。方法:用高效液相色谱法(HPLC)测定血浆5-Fu浓度。结果:犬皮下植入5-FuI20mg/kg后血药浓度—时间变化过程符合具有缓释的一房室模型,缓释半衰期T0.5r=52.7±10.9h,在体内维持有效浓度(≥1ug/mL)时间长达216~240h。结论:5-FuI皮下植入确具缓释作用和维持有效浓度时间长的优点。Objective:To investigate the pharmacokinetics of 5-Fu implant (5-FuI) in dogs.Methods:The 5-FuI concentration in plasma were detected with HPLC.Results:It was found that the characteristics of the plasma drug concentration\|time curve fit the one compartment open model with sustained release preparation after subcutaneous embedding of 5-FuI 20mg.kg -1 to dogs.The sustained release half\|life time (T 0.5 r)was as long as 52.7±10.9hrs.The 5-Fu concentration in plasma was higher than the effective anticancer concentration (1 μg.mL -1 ) for 216~240hrs.Conclusions:5-Ful embedded subcutaneously has markedly sustained releasing effect,5-FuI is a new dosage form for 5-Fu with excellently pharmacokinetic characteristics.
