摘  要:目的:通过观察滋阴活血解毒方对局灶性脑缺血模型大鼠血清及脑组织炎性因子表达的影响,探讨该方对脑缺血大鼠炎症反应的干预作用。方法:将32只大鼠随机分为假手术组、模型组、中药组、阿加曲班组,每组8只。对除假手术组外的其他3组进行大脑中动脉阻塞(MCAO)造模,造模成功2h后给药,治疗组给予滋阴活血解毒方灌胃,对照组给予阿加曲班腹腔注射,假手术组和模型组均灌服等剂量生理盐水,造模后24h,麻醉大鼠,检测各组大鼠血清中IL-1β、IL-8、TNF-α的含量,缺血部位脑组织MCP-1、NF-kBp65的表达。结果:模型组大鼠血清IL-1β、IL-8、TNF-α含量,脑组织中MCP-1、NF-kBp65表达明显高于假手术组(P<0.05,P<0.01),中药组、阿加曲班组均能降低大鼠血清IL-1β、IL-8、TNF-α含量,降低MCP-1、NF-kBp65的表达。结论:滋阴活血解毒方能通过抑制炎症因子的表达,减轻脑缺血后炎症反应,从而发挥脑保护作用。Objective : To observe the effect of yin - nourishing, blood circulation - activating, and detoxicating prescription on the expression of inflammatory factors in serum and brain tissue in the rat model of focal cerebral ischemia and to investigate the interven- tional effect of the prescription on the inflammatory response in rats with cerebral ischemia. Methods:Thirty - two rats were randomly and equally divided into sham - operation group, model group, traditional Chinese medicine ( TCM ) group, and argatroban group. All groups except the sham - operation group underwent middle cerebral artery occlusion to establish a rat model of cerebral ischemia. At 2 h after the model was established,the TCM group received yin -nourishing, blood circulation -activating, and detoxicating prescription by gavage,the argatroban group received argatroban by intraperitoneal injection, and the sham- operation group and model group were orally given an equal volume of saline. At 24 h after the model was established, the rats were anesthetized;the serum interleukin (IL) - 1β, IL - 8, and tumor necrosis factor (TNF) - α levels and the expression of monocyte chemotactic protein (MCP) - 1 and nuclear factor - kappa B p65 ( NF -κB p65 ) in ischemic brain tissue were measured. Results : Compared with the sham - operation group, the model group had significantly higher serum IL - 1β,IL -8,and TNF - α levels and expression of MCP - 1 and NF -κB p65 (P 〈0. 05 or P 〈 0. 01 ). The TCM group and argatroban group showed decreases in serum IL - 1β, IL - 8, and TNF - ot levels and expression of MCP- 1 and NF-κB p65. Conclusion:Yin -nourishing, blood circulation- activating, and detoxicating prescription can protect the brain by inhibiting the expression of inflammatory factors and reducing inflammatory response to cerebral ischemia.
