摘 要:2012年初从东台地区分离到3株H9亚型禽流感病毒,通过RT-PCR扩增出其HA基因,并对其进行测序及遗传进化分析。结果显示3株病毒的HA基因开放阅读框全长1683bp,共编码560个氨基酸,其核苷酸和氨基酸的同源性分别为95.8%~97.8%、97.7%~98.9%,与A/chicken/F/98的同源性分别为90.9%~91.4%、93.2%~93.9%;其裂解位点均为PSRSSR↓GLF,符合低致病力禽流感毒株的特征,推导的氨基酸序列含8个相同的潜在糖基化位点,A/chicken/DT/1/2012比其他2株H9亚型AIV多了一个267NCT的糖基化位点;受体结合位点的226位为L,具有α-2,6受体结合的特征。遗传进化分析显示3株分离株属欧亚种系,与A/Duck/Hong Kong/Y280/97代表株的亲源关系较近。The hemagglutinin (HA)genes of three H9 subtype avian influenza viruses (AIVs)isolated in Dongtai area in 2012 were amplified by RT-PCR and their sequences were analysed. The results showed that HA genes of AIV isolates were composed of 1 683 bp nueleotides, encoding 560 amino acids. Nucleotide and amino acid homologies among these AIV isolates were from 95.8% to 97.8% and 97.7% to 98.9%,respectively. They shared 90.9% to 91.4% and 93.2% to 93.9% homologies with A/chicken/F/98 strain. The cleavage site of HA proteins was PSRSSR ↓ GLF,which had the characteristic of low pathogenic AIV strain. There were eight potential glycosylation sites in the HA genes of three isolates, and A/chicken/DT/1/2012 strain had an additional glycosylation site at position 267. The Leucine at position 226 in the HA genes of 3 isolates indicated the potential of binding with SA α,2-6 receptor. Phylogenetic analysis of HA genes showed that 3 isolates belonged to Eurasian lines,which had a close relationship with A/Duck/ Hong Kong/Y280/97 AIV strain.
