六味地黄丸提取物对小鼠支持细胞体外增殖的影响

摘　　要：目的探讨六味地黄丸提取物对小鼠支持细胞体外增殖的影响。方法通过形态学观察、MTT法检测细胞活力、TUNNEL法检测细胞凋亡指数和Brdu标记法检测细胞增殖活性等研究5个不同浓度的六味地黄丸提取物对小鼠支持细胞体外生长和增殖的作用。结果当用20 mg/L以上的六味地黄丸提取物处理培养的小鼠支持细胞时,TENNEL法获得的细胞凋亡指数、MTT法测得的细胞生长抑制率和Brdu标记法测得的细胞增殖率与对照组相比有显著差异,并呈剂量依赖效应;细胞形态观察结果也证实这一现象。结论六味地黄丸提取物以剂量依赖方式促进体外培养的小鼠支持细胞的增殖,抑制细胞凋亡,可能通过促进支持细胞的增殖与活性促进精子发生。In the present study,the effects of extracts of Liuwei dihuang pill（LW） with 5 of different concentration on the cultured Sertoli cells in DMEM medium were investigated by using the morphological observation,MTT supravital staining for cell viability assessment,Tunnel method for cell apoptosis and BrdU labeling for cell proliferation monitor.The results indicated that when the cultured Sertoli cells were treated with 10 mg/L or more of extracts of LW,the apoptotic index from TENNEL method had no significantly differences compared with the data of the control,inhibitory rate of Sertoli cells measured by MTT method,and the data of cell proliferation monitor by BrdU labeling all showed the significantly differences compared with the data of the control with a dose-dependent manner,and the morphological observation also revealed the above phenomena.Therefore,the extracts of LW enhances enhanced the growth and proliferation of cultured Sertoli cells in vitro,which probably promote the spermatogenesis.
