摘　　要：目的探讨电针对大鼠坐骨神经损伤后神经导向因子Netrin-1及其mRNA表达的影响。方法健康SD大鼠72只,随机分为空白对照组、模型对照组和电针治疗组,每组24只。后两组大鼠右坐骨神经横断后即刻端对端缝合。电针治疗组取"环跳"、"足三里"进行电针治疗,1次/d,7天1个疗程,连续治疗3个疗程。每疗程结束后应用免疫组化法、逆转录-聚合酶链反应(RT-PCR)分别检测坐骨神经和相应脊髓段Netrin-1及其mRNA的表达变化。结果电针组与模型组比较,大鼠损伤的坐骨神经和相应脊髓段中Netrin-1及其mRNA在第一疗程后达到高峰,之后逐渐降低,且都高于模型组,有非常显著性差异(P<0.01),而且两组始终都高于空白对照组,也有非常显著性差异(P<0.01)。结论电针治疗能明显增强损伤的坐骨神经和相应脊髓段中Netrin-1及其mRNA的表达。Objective Purpose To approach to the effects on nerve growth guidance cue Netrin-1 of rats after sciatic nerve injury.Methods 72 healthy SD rats were randomly divided into control group,model group and the electroacupuncture（EA） group（n = 24）.In the latter two groups,the right sciatic nerve was transected and end-to-end suture immediately.The treatment of EA on ＂ Huantiao＂（GB 30） and＂ Zusanli＂（ST 36） was applied once daily,and 7 days a course of treatment,continuous treatment of three courses.The changes in expression of Netrin-1 and Netrin-1mRNA in sciatic nerve and corresponding section of spinal cord were detected by RT-PCR and immunohistochemical methods after the end of each course of treatment.Results EA group compared with model group,Netrin-1 and its mRNA in the injured rat sciatic nerve and spinal cord segments reached a peak after the first course of treatment,then decreased gradually,and are higher than the model group,there was a remarkably significant difference（P 0.01）,and the two groups is always higher than the control group,there are also remarkably significant differences（P 0.01）.Conclusion EA treatment can remarkably strengthen expression of Netrin-1of slit1 and Netrin 1mRNA in sciatic nerve and corresponding section of spinal cord after sciatic nerve injury.
