摘  要:目的研究哺乳期多溴联苯醚-153(BDE-153)暴露对成年大鼠海马神经细胞凋亡及p53基因和蛋白表达的影响。方法将40只4日龄清洁级SD雄性大鼠按体重随机分成4组,分别为对照(橄榄油)组和1、5、10 mg/kg BDE-153染毒组,每组10只。于出生第10天(PND10),采用腹腔一次性注射方式进行染毒,染毒容量为10 ml/kg。染毒2个月后,测定海马神经细胞的凋亡率和TUNEL阳性细胞率及海马p53基因和蛋白的表达。结果与对照组相比,10 mg/kg BDE-153染毒组大鼠海马神经细胞TUNEL阳性细胞率和p53基因的表达均升高,5、10 mg/kg BDE-153染毒组大鼠海马神经细胞凋亡率和p53蛋白的阳性率均增高,差异有统计学意义(P<0.05)。且随着BDE-153染毒剂量的升高,大鼠海马神经细胞凋亡率和TUNEL阳性细胞率及p53基因的表达和p53蛋白的阳性率均呈上升趋势。结论 p53基因和蛋白可能参与了BDE-153诱导的神经细胞凋亡过程。Objective To study the effects of polybrominated diphenyl ether-153 (BDE-153) exposure during lactation on the hippocampus cell apoptosis and p53 expression in adult male rats,and to provide the scientific basis for BDE-153 neurotoxicity. Methods Forty newborn clean grade Sprague Dawley male rats were randomly divided into four groups at postnatal day 4 (PND4) according to their body weights, 1,5,10 mg/kg body weight of BDE-153 groups and olive oil solvent control group, 10 in each group. At PND10,rat pups were administrated BDE-153 olive oil solution by intraperitoneal injection at 10 ml/kg body weight for one time. Two months after treatment,the hippocampus cell apoptosis and p53 gene and protein expressions were detected. Results Compared to the controls,the rats' hippocampus p53 gene expression and TUNEL positive cell rates significantly increased in 10 mg/kg BDE-153 group,p53 protein positive rates and neuron apoptosis rates significantly increased in the 5,10 mg/kg BDE-153 groups compared with the controls (P〈0.05). Furthermore,the rats' hippocampus neuron apoptosis rates,TUNEL positive cell rates,p53 gene expressions and p53 protein positive rates showed rising trends with the increasing BDE-153 exposure levels. Conclusion p53 gene and protein may be involved in the process of BDE-153-induced hippocampus cell apoptosis.
