摘  要:在真核生物的细胞核内 ,基因组是通过 DNA的核骨架附着区 ( SAR)或称核基质附着区( MAR) (简记为 S/MAR)锚定在核骨架网状系统上的 .S/MAR既有一定的特征 ,又有多样性 ,研究认为它参与了 DNA复制调控和转录调控等多种核内生化过程 .通过重组 ,在目的基因一侧或两侧带上 S/MAR后作基因转染或转基因动植物 ,发现整合后的基因表达有时可增强几倍 ,甚至上万倍 ,和 /或显示位置独立效应 .有些研究还报道 ,S/MAR能克服转基因在传代过程中存在的转基因沉默的问题 .这些功效使得 S/MAR将可能成为基因工程中一个有用的顺式调控元件 .本文报道了关于Within eukaryotic nuclei, the genomic DNA is organized into topologically constrained domains by DNA regions (called scaffold attachment regions [SARs], or matrix attachment regions [MARs]) anchoring chromatin to the matrix. All S/MARs have several features in common, but they are various. Some studies found that some S/MARs were associated with many biochemical processes such as the regulation of DNA replication and transcription. Some experiments of gene transfection or of transgene to animal or plant found that when the foreign gene was flanked by S/MAR on one side or both sides by gene recombination, the expression of the integrated foreign gene could be increased to several fold, sometimes even to 10000 fold, and/or was independent of chromosomal position. In some instances, it has been reported that S/MAR has the function to decease the transgene silencing. All of these effects of S/MAR would make it becomes a new type of cis acting DNA element which is very useful for increasing transcription activity and ensuring the correct control of randomly inserted transgenes in gene engineering. This paper reported the development of the research about the relationship between S/MAR and gene expression ,and some instances of the utilization of S/MAR.
