摘 要:目的研究槲皮素对过氧化氢损伤的大鼠胸主动脉内皮细胞的保护作用。方法体外培养大鼠胸主动脉内皮细胞,用过氧化氢诱导血管内皮细胞损伤,观察槲皮素对大鼠胸主动脉内皮细胞存活率的影响,测定培养液中NO、MDA、SOD的水平。结果过氧化氢对血管内皮细胞具有损伤作用,20~80μmol/L的槲皮素可浓度依赖性保护血管内皮细胞,升高细胞培养液中NO水平,降低MDA含量,提高SOD活力。结论槲皮素可保护过氧化氢损伤的血管内皮细胞,其保护作用与促进血管内皮细胞释放NO、抑制脂质过氧化以及抗氧化有关。Objective To study the protective effects of quercetin against H2 O2-induced injury of rat aorta endothelial cells (RAECs). Methods RAECs were cultured in vitro and the cells were injured by H2 02, the cells viability were observed and the levels of NO, MDA and SOD in the culture medium were measured. Results RAECs were damaged obviously by H2O2. Quercetin (20-80 μmol/L) protected RAECs in a concentration-dependent manner. The level of NO and the activity of SOD were increased, and the level of MDA were decreased. Conclusion Quercetin has protective effect against RAECs injuries induced by H2O2. The mechanism of quereetin' s cyto- protective effect may be related to its stimulation of NO release from endothelial cells and anti-lipid peroxidation.
