摘  要:目的:研究大豆甙元(Daidzein,Da)联合TRAIL对SGC-7901细胞的影响。方法:采用不同浓度的Da和TRAIL单独及联合作用于SGC-7901细胞,通过MTT细胞和流式细胞术检测增殖、凋亡和周期的变化情况,透射电镜观察对细胞内部形态和微结构的影响,利用Western blot检测对相关凋亡基因Bcl-2、survivin、caspase-3的表达变化。结果:Da联合TRAIL处理人胃癌SGC-7901细胞后,生长有明显的抑制作用,并且具有一定的剂量依赖性;细胞周期阻滞于S期;细胞内部出现典型凋亡形态学改变;Bcl-2和survivin蛋白表达下调,caspase-3蛋白被上调。结论:Da联合TRAIL通过细胞周期S期阻滞,及下调Bcl-2、survivin蛋白及增强caspase3蛋白表达而诱导胃癌细胞凋亡。Objective:To investigate the effects of Daidzein combined with TRAIL in gastric adenocarcinoma cell SGC- 7901. Methods: SGC-7901 cells were treated with Da or/and TRAIL at different concentration for indicated times. MTY assays were per- formed to measure the growth inhibitory effect of Da or/andTRAIL. Cell cycle and apoptosis of cells were analyzed using flow cytome- try. Transmission electron microscope was introduced to Observe the changes of internal morphology and microstructure of SGC-7901 cells. Western blot was used to examine the changes of Bcl-2, caspase-3, survivin expression. Results: Da combined with TRAIL obvi- ously inhibits the cell growth of SGC-7901 cells in a doseTdependent manner and induces cell cycle arrest in S phase. Typical apoptosis morphology changes were observed after the combined treatment. Bcl-2 and survivin proteins were decreased and the level of caspase-3 was increased upon the combined treatment. Conclusion:Da combined with TRAIL lead to apoptosis of gastric adenocarcinoma cells through induction of cell cycle arrest in S phase, inhibition.of Bcl-2 and survivin protein expression, and enhancement of caspase-3 protein level.
