摘 要:目的:研究低浓度乙醇对铜绿假单胞菌磷脂酶C基因(PLCH)表达的影响。方法:观察不同浓度乙醇对铜绿假单胞菌生长曲线影响,荧光定量实时PCR检测不同乙醇浓度培养的铜绿假单胞菌PA01的PLCH表达量变化。、结果:工程株PA01在0.1%、0.5%、1%乙醇浓度下,其生长曲线与对照基本一致,在孵育18h都达到最高生长密度;在2%~5%的乙醇浓度下,细菌生长极其缓慢,6%以上的乙醇浓度下几乎不生长..与对照相比,在0.1%、0.5%和1%乙醇诱导下.PLCH的表达量的分别比对照组上升了44倍、30倍和8倍.在2%乙醇浓度下,PLCH表达量略为下降。结论:低浓度乙醇有助于铜绿假单胞菌相关毒力因子表达上调,增加生存力.在感控消毒中,保持足够的酒精浓度对清除铜绿假单胞菌至关重要。Objective:To study the effects of low concentration of ethanol on phospholipase C (PLC) gene expression of Pseudomonas aeruginosa. Methods : The effects of different concentrations of ethanol on the growth curve of Pseudomonas aeruginosa (P. aeruginosa) were determined. The change in PLCH expression of PAO1 strain incubated with different concentrations of ethanol was assayed via real-time polymerase chain reaction. Results: Compared with control group, the PAO1 strain yielded a similar growth trend when incubated with 0. 1%, 0.5%, and 1% ethanol, with the density (OD600) of the strain culminated at 18 h. The PAO1 strain had a retarded growth when incubated with 2% to 5% ethanol and ceased to grow when incubated with 〉 6% ethanol. The PLC expression of P. aeruginosa was increased by 44-, 30-, and 8-fold when incubated with 0. 1%, 0.5% and 1% ethanol, respectively, compared with control group. Conversely, PLC gene expression was slightly attenuated by 2% ethanol. Conclusion: Low concentrations of ethanol may promote the survival of P. aeruginosa via up-regulation of the associated virulence factors. Disinfection using ethanol at a sufficient concentration is crucial to clear P. aeruginosa.
