靶向Dicer酶的小干扰RNA对人脑胶质瘤细胞生物学行为的影响

摘　　要：目的观察敲低Dicer酶的表达对人脑胶质瘤细胞U87、U251的生物学行为的影响。方法采用小干扰RNA（siRNA）技术敲低Dicer酶的表达，Westernblot检测U87、U251细胞转染小干扰RNA24h后Dicer酶表达；应用细胞计数试剂盒（CCK-8）法检测、Tranwell试验及明胶酶谱实验检测细胞增殖、侵袭及基质金属蛋白激酶[基质金属蛋白酶（MMP）-2、MMP-9]的活性。结果转染siRNA靶向Dicer酶后，Dicer酶较对照组降低68．0％（U87）和60．3％（U251）（P〈0．05）。两组细胞的增殖率分别明显高于相对应的对照组，且随时间的延长差距也随之增大（P〈0．05）。siRNA组细胞侵袭数（77．9±8．3）个（U87）和（81．0±7．9）个（U251）较阴性对照组（40．6±7．0）个（U87）和（35．8±6．5）个（U251）明显增多（P〈0．05）。siRNA组MMP-2较对照组活性增加（1．35±0．17）倍（U87）、（1．19±0．06）倍（U251）（P〈0．05）；MMP-9较对照组活性增加（1．72±0．37）倍（U87）、（1．66±0．33）倍（U251）（P〈0．05）。结论敲低Dicer酶表达后，人胶质瘤细胞增殖能力增加，MMP-2、MMP-9活性增强，侵袭性增强。Objective To observe the effects of knocking down Dicer enzyme expression on the bi- ological behaviors of human glioma cells U87 and U251. Methods Using small interference RNA （siRNA） technology, Dicer enzyme expression was knocked down , Western blot detection U87 and U251 cells＇ Dicer enzyme expression after transfected with small interfering RNA 24 h ; application of cell counting kit-8 （CCK-8） assay, Tranwe]l test and gelatinase spectrum experiments to detect cell proliferation, invasion and matrix metalloproteinase [ matrix metalloproteinase （MMP） -2, MMP-9 ] activity. Results Transfected with siRNA targeting Dicer enzyme, Dicer enzyme compared with the control group decreased by 68.0% （ U87 ） and 60. 3% （ P 〈 O. 05 ） （ U251 ）. Both cell proliferation rate was significantly higher than the cor- responding control group, and with time gap also increased （ P 〈 O. 05 ）. number of cell invasion siRNA group 77.9_±8.3 （U87） and 81.0 ±7.9 （U251） compared with negative control group 40.6 -±7.0 （ U87 ） and 35.8 -± 6. 5 （ U251 ） increased significantly （ P 〈 0. 05 ）. The siRNA set of MMP-2 activity in- creased compared with the control group （ 1.35 ± 0. 17） times （ U87）, （ 1.19 ± 0. 06） times （U251） （ P 〈 0. 05 ） ; MMP-9 activity increased compared with the control group （ 1.72 ± 0. 37 ） times （ U87 ）, （1.66 ±0.33） times （U251） （P〈0.05）. Conclusion After dicer enzyme expression knockdown,hu- man glioma cell proliferation increased, MMP-2, MMP-9 activity was enhanced and invasive increased.
