泽泻汤加味方治疗高血压最佳组方配伍的正交试验研究

摘　　要：目的探讨泽泻汤加味方降压效果的最佳组方配伍规律。方法取造模成功的高血压模型大鼠54只,随机分为9组。根据泽泻汤原方宗旨,确定泽泻(因素A)为君药;并以原方的用量比例泽泻∶白术=5∶2为基础,确定白术(因素B)的相应剂量;考虑到加用石菖蒲、泽兰(因素C)后的影响,确定石菖蒲、泽兰2味用量相等,2味用量之和不超过泽泻汤原方用量,设计为3因素3水平正交试验。各组按正交表中相对应的剂量配伍而成的水煎剂进行灌胃给药,连续4周。测定实验前后大鼠尾动脉血压及尿量变化。结果泽泻汤加味方降压效果最佳组合为:A1B1C3,即泽泻21 g,白术9 g,泽兰、石菖蒲各15 g。结论泽泻汤加味方在泽泻∶白术=5∶2,且泽泻+白术=泽兰+石昌蒲的药用剂量时,降压效果最佳;其降压作用初期与利尿有关,长期降压作用与利水活血祛痰的综合作用相关。Objective To explore the law of the optimal ingredient composition of modified zexietang for the anti - hypertensive effect. Methods Fifty - four rats of successfully modeled hypertension were collected and randomized into 9 groups. According to the therapeutic actions of the original zexietang, zexie [ （ rhizoma alismatis）, Factor A 1 was determined as the monarch herb. Based on the dosage ratio in the original formula,the ratio of zexie（ rhizoma alismatis）and baizhu（ rhizoma atractylodis macrocephalae） = 5：2 ,the corresponding dose of baizhu [ （rhizoma atractylodis macrocephalae）, Factor B ] was determined. In consideration of the impacts of the supplementary herbs, shichangpu and zelan [ （ rhizoma acori graminel） （ herba ly- copi ） , Factor C ） 1 , it was to determine that the doses of these two herbs were equal and the total dose of these two herbs should be not exceeded the dose of the original formula. Finally, the orthogonal design trial with three factors and three levels was set up. The decoction was prepared according to the composition of corresponding herbal doses in the orthogonal array in each group. The lavage was done continuously for 4 weeks. The changes in the tail arterial blood pressure and urine volume in the rats before and after treatment were determined. Results The optimal ingredient composition of modified zexietang ： A1B1C3 , i. e. zexie （ rhizoma alismatis ） 21 g, baizhu （ rhizoma atractylodis macrocephalae） 9 g, shichangpu and zelan [ （ rhizoma acori graminel）, （ herba lycopi） ] 15 g for each. Conclusion The effect of modified zexietang on hypertension is optimal in the dosage of zexie （ rhizoma alismatis） ： baizhu （ rhizoma atractylodis macrocephalae） = 5 ： 2, and the dosage of zexie（rhizoma alismatis） ＋ baizhu（ rhizoma atractylodis macrocephalae） = the dosage of shichangpu（ rhizoma acori graminel）and zelan（ herba lycopi）.
