摘  要:目的探讨人胰高糖素样肽1(GLP-1)对非肥胖型糖尿病(NOD)小鼠胰腺导管上皮细胞分化与增殖的影响。方法 GLP-1治疗组小鼠用微型渗透泵皮下持续泵入人GLP-1,对照组小鼠泵入生理盐水,4周后将其胰腺组织分别做导管细胞角蛋白(DCK)/胰岛素双重免疫荧光染色、胰腺十二指肠同源异型盒基因(PDX-1)/胰岛素双重免疫荧光染色及5-溴脱氧尿嘧啶核苷(BrdU)/DCK双重免疫染色,显微镜下观察胰腺导管上皮细胞的分化和增殖。结果在GLP-1治疗组胰腺导管上皮中可见较多胰岛素阳性细胞及PDX-1/胰岛素双阳性细胞,而在对照组几乎看不到;GLP-1治疗组小鼠BrdU阳性的导管上皮细胞的百分数与对照组小鼠相比明显增高(P<0.001)。结论人GLP-1持续刺激NOD小鼠后,可促进胰腺导管上皮细胞分化与增殖。【Objective】 To To investigate the effect of continuous stimulation of human glucagon-like peptide-1(GLP-1) on differentiation and proliferation of pancreatic duct epithelium in Non-obese diabetic(NOD) mouse.【Methods】 The mice in the GLP-1 group and the control group were infused subcutaneously with human GLP-1 and saline respectively via a mini-osmotic pump for 4 weeks.Duct epithelium differentiation and proliferation were examined by duct cytokeratin(DCK)/insulin,pancreatic and duodenal homeobox gene-1(PDX-1)/insulin double immunofluorescence staining and 5-bromodeoxyuridine(BrdU)/DCK double immunostaining respectively.【Results】 There were more insulin-positive cells and PDX-1/insulin double-positive cells in duct epithelium in the GLP-1 group,which were rarely detected in the control group.The percentage of BrdU-positive duct epithelium in the GLP-1 group was significantly higher than that in control group(P &lt;0.001).【Conclusion】 Continuous infusion of human GLP-1 to prediabetic NOD mouse induces pancreatic duct epithelium differentiation and proliferation.
