摘 要:[目的]研究穿山龙总皂苷对哮喘小鼠气道重塑和平滑肌肌动蛋白(α-SMA)的影响。[方法]SPF级BALB/c小鼠50只随机分为空白组、模型组、穿山龙总皂苷高剂量组、中剂量组、低剂量组。卵蛋白(OVA)致敏激发小鼠制作哮喘气道重塑模型,在实验第18-55d,空白组、模型组以生理盐水灌胃,药物组分别给予穿山龙总皂苷80mg.kg-1、40 mg.kg-1、20 mg.kg-1治疗。实验结束后HE染色观察哮喘小鼠肺组织病理形态变化,采用免疫组织化学法检测α-SMA水平。[结果]穿山龙总皂苷能改善哮喘小鼠气道壁及支气管平滑肌的增厚;与空白组比较,模型组小鼠α-SMA明显升高(P<0.05);与模型组比较,高、中剂量组α-SMA水平明显降低(P<0.05)。[结论]穿山龙总皂苷能够抑制哮喘小鼠肺组织表达α-SMA,影响平滑肌增厚,改善哮喘小鼠气道重塑状态。[Objective]To Study the effect of Con-saponin of Dioscoreae Nipponicae(CDN) on expressions of airway remodeling and smooth muscle actin-α(α-SMA)in asthmatic mice.[Method] Fifty SPF BALB/c mice were randomly divided into normal group,model group,high-dose CDN group,medium-dose CDN group,low-dose CDN group.The five groups except normal group were induced as asthmatic modeling allergies mice with ovalbumin(OVA).During the 18~55 days in the experiment,the normal and model groups were treated with normal saline.Meanwhile,CDN groups were respectively treated with 80 mg.kg-1,40 mg.kg-1,20 mg.kg-1 of CDN.At the end of experiment,we observed the path-morphological changes in lung issues and took the sample blood to detect α-SMA using IC.[Result]The pathological changes in lung tissue showed that CDN could significantly inhibit the thickening of airway well and bronchial smooth muscle.The level of α-SMA was significantly increased except the normal group(P&lt;0.05).In highdose and medium-dose CDN groups,the level of α-SMA was significantly decreased of α-SMA which reduced bronchial smooth muscle thickening and suppressed airway remodeling in asthmatic mice.
