HPLC法测定单面针药材中白鲜碱含量

摘　　要：本文成功地建立了一种测定单面针药材中白鲜碱含量的高效液相色谱(HPLC)分析方法。样品采用超声波辅助提取,色谱柱为SinoChrom ODS-BP C18柱(4.6 mm×250 mm,5μm),以乙腈-0.01 mol/L磷酸二氢钠缓冲液(50:50)为流动相,流速0.8 mL/min,在波长236 nm处进行白鲜碱含量检测。实验结果表明,白鲜碱在进样量为0.001 5～0.200 0μg范围内,峰面积和质量浓度呈良好的线性关系(R2=0.9995);平均回收率为98.8%;相对标准偏差(RSD)为3.06%(n=6)。该法准确可靠、重现性好,可用于对不同产地、不同药用部位单面针中白鲜碱含量的测定。An analysis method using high performance liquid chromatographic for determining dictamnine in Zanthoxylum dissitum Hemsl had been successfully established.In the experiments,dictamnine was obtained by ultrasonic extraction and separated on SinoChrom ODS-BP C18 column（4.6 mm × 250 mm,5 μm） and the mobile phase was mixture liquid of acetonitrile and 0.01 mol/ L NaH2PO3（50 ︰ 50,V/V）;the flowing speed of the mobile phase was 0.8 mL/min,and the wavelength for detecting dictamnine was 236 nm.The results indicate that when the injection volume of the dictamnine ranged from 0.0015 to 0.200 0 μg,a significant linear relationship showed between the peak area and the mass concentration（R2=0.9995）,the average recovery rate of dictamnine was 98.8%,and the relative standard deviation（RSD） was 3.06%（n=6）.In conclusion,the established analysis method possessed good accuracy,reliability and high repeatability,which made it a suitable detection method for determining dictamnine in Z.dissitum from different origin place and medicinal parts.
