摘 要:目的观察紫金砂(Angelica polymorpha Maxim.)对家兔心肌缺血再灌注损伤(MIRI)的保护作用。方法 56只新西兰大耳白兔随机分为7组:假手术组、缺血再灌注模型组、复方丹参滴丸(1.0 g/kg)组、维拉帕米(0.2 mg/kg)组、紫金砂(0.25、0.5和1.0 g/kg)组。采用结扎冠状动脉左前降支30 min,再灌注120 min的方法建立家兔MIRI模型。复方丹参滴丸组及紫金砂各剂量组实验前灌胃给药7 d;维拉帕米组于缺血前10 min采用蠕动泵股静脉一次性滴注给药。记录手术前后不同时间点心电图;2,3,5-氯化三苯基四氮唑(TTC)染色方法测定心肌梗死面积;比色法检测血清肌酸激酶(CK)、乳酸脱氢酶(LDH)、超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量。结果与模型组比较,各给药组动物异常升高的T波及S-T段明显降低,心肌梗死面积明显减小(P<0.01);紫金砂各给药剂量组组间比较,紫金砂0.5及1.0 g/kg给药组动物心肌梗死面积明显低于紫金砂0.25 g/kg给药组(P<0.01),但复方丹参滴丸组、维拉帕米组、紫金砂0.5及1.0 g/kg给药组组间比较无显著差异。与模型组比较,各给药组CK、LDH及MDA值明显降低(P<0.01),且SOD活性明显升高(P<0.01);各给药组组间比较,除紫金砂1.0 g/kg给药组动物血清CK值明显低于紫金砂0.25 g/kg组外(P<0.05),其余各给药组血清学指标(CK,LDH,SOD活性及MDA含量)组间比较均无统计学意义。结论紫金砂对MIRI具有明显保护作用,其作用机制可能与其抗氧化作用有关。Objective To observe the protective effect of Angelica polymorpha Maxim.on the myocardial ischemia-reperfusion injury(MIRI) in rabbits.Methods Fifty-six New Zealand white rabbits were randomly divided into 7 groups: sham operation group,ischemia-reperfusion(I / R) model group,Compound Danshen dripping pill group(1.0 g / kg),verapamil group(0.2 mg / kg),as well as 0.25,0.5 and 1.0 g / kg Angelica polymorpha Maxim.groups.The MIRI models in rabbits were induced by ligation of the left anterior descending coronary artery(LAD) for 30 min followed by reperfusion for 120 min.Before the experiment,animals received daily compound Danshen dripping pills and Angelica polymorpha Maxim.for 7 days by intragastric administration;and the verapamil group were administrated by femoral vein injection through peristaltic pump within 10 min at constant speed for 10 min before ischemia.The electrocardiogram was recorded at different time points since ischemia.The myocardial infarct size was determined by 2,3,5-triphenyl-2Htetrazolium chloride(TTC) staining.The creatine kinase(CK),lactate dehydrogenase(LDH),and superoxide dismutase(SOD) activities,as well as the malondialdehyde(MDA) content in serum were analyzed by colorimetric technique.Results Compared with the I / R model group,the T wave,S-T segment,and the myocardial infarct sizes of all the administrated groups were significantly reduced(P 0.01).In addition,the myocardial infarct sizes of 0.5 and 1.0 g/kg Angelica polymorpha Maxim.treated group were lower than those of 0.25 g / kg Angelica polymorpha Maxim.treated group(P 0.01).But there was no remarkable difference in myocardial infarct size among Compound Danshen dripping pill,verapamil,and 0.5 and 1.0 g / kg Angelica polymorpha Maxim groups.Compared with the I / R model group,the CK,LDH and MDA values of all the administrated groups were significantly decreased(P 0.01),but the activity of SOD was increased(P 0.01).In addition,the CK value of 1.0 g / kg Angelica polymor
