五味子含药血清中药源性成分的HPLC-DAD分析

摘　　要：目的分析五味子含药血清中的药源性成分。方法大鼠灌胃给予五味子乙醇提取物制备含药血清,HPLC-DAD测定含药血清、五味子提取物、对照品及空白血清指纹图谱,各色谱峰紫外光谱图,通过色谱峰保留时间和光谱图两方面比对,分析大鼠给药后体内产生的药源性成分。结果含药血清中产生药源性成分13个,其中8个为药材原形成分入血,5个为代谢产物,8个原形成分中的4个分别为五味子醇甲、五味子醇乙、五味子甲素和五味子乙素,所有13个药源性成分均具有联苯环辛烯类木脂素的吸收特征。结论五味子含药血清中的药源性成分主要为联苯环辛烯类木脂素及其代谢产物。Objective To analyze drug-induced constituents in serum containing drug （SCD） of Schisandra chinensis （SC）. Methods SCD were prepared after rats of gavage administration of ethanol extract of SC. Chromatogram fingerprint of SCD, ethanol extract of SC, reference substance and blank serum were detected by HPLC-DAD and spectrograms of chromatographic peaks of these chromatograms were extracted, respectively. By comparing the retention time and spectrograms of chromatographic peaks, drug-induced constituents in rats serum were analyzed. Results Thirteen drug-induced constituents were detected in rats after oral administration of SC, eight of them were the original compounds contained in materia medica and five were metabolites, and four constituents out of eight original form ones were identified as schisandrin, schisandrol B, deoxyschisandrin, and schisandrin B. All the spectrograms of thirteen drug-induced constituents possessed the same character as dibenzocyclooctadiene-like products. Conclusion The main drug-induced constituents in SCD of SC were dibenzocyclooctadienes and its metabolites.
