摘  要:目的研究潜阳育阴颗粒对SHR血液及肾脏AngII含量及肾脏TNF-α表达的影响。方法 40只14周龄雄性SHR随机分为模型组(等量蒸馏水)、缬沙坦组(0.027 g/kg/d)、潜阳育阴颗粒低、中、高剂量组(2.5 g/kg/d、5 g/kg/d、10 g/kg/d),每组8只;8只同周龄雄性Wister大鼠为正常组(等量蒸馏水)。灌胃给药,每日1次,连续6周。末次给药1 h后,取大鼠血清及肾脏。无创性尾动脉测压法检测动脉压,全自动生化仪和放免法检测尿m-ALB、尿β2-MG、尿NAG,ELISA法检测血液和肾脏AngII,免疫组织化学法观察肾脏TNF-α表达。结果潜阳育阴颗粒可以降低SHR动脉压、尿m-ALB、尿NAG;SHR血液及肾脏中AngII含量较Wister明显升高,潜阳育阴颗粒显著降低SHR血液及肾脏AngII含量;SHR肾脏TNF-α表达较Wister明显增多,潜阳育阴颗粒显著降低TNF-α的表达。结论潜阳育阴颗粒对高血压及其早期肾脏损害具有一定的防治作用,可能与其降低AngII含量及TNF-α表达有关。Objective To investigate the effect of QianYangYuYin (QYYY) granules on the levels of Angiotensin Ⅱ (AngⅡ) in the blood and renal tissue and the expression of tumor necrosis factor-α (TNF-α) in the renal tissue of Spontaneously Hypertensive Rats (SHR). Methods 40 male SHR of 14 weeks aged were randomly divided into the model group (the same amount of distilled water), the group of valsartan (0.027 g/kg/d), the low dose group (2.5 g/kg/d), the medium dose group (Sg/kg/d) and the high dose group (10 g/kg/d) of QYYY granules with 8 SHR each group, and 8 male Wister rats of the same aged as the control group (the same amount of distilled water) were chosen. Rats were filled into the stomach with different medicines according to the different groups once daily for 6 weeks. The kidneys and blood serum were taken out 1 hour later after the last administration in rats of each group. The arterial pressure was tested by the non-invasive measurement of blood pressure by caudal artery of rats. Urinary micro-albumin, urinary β2-micro-globulin and urinary N-acetyl-glucosaminidase were determined by full automatic biochemical analyzer and radioimmunoassay. The levels of AngII in the blood and renal tissue of SHR were determined by enzyme linked immune-sorbent assay. The expression of TNF-α in the renal tissue of SHR were tested by immunohistochemical method. Results QYYY granules could reduce arterial pressure, urinary micro-albumin and urinary N-acetyl-glucosaminidase of SHR significantly. The levels of AngII in the blood and renal tissue of SHR were higher than those of Wisters. QYYY granules could decrease the content of AngⅡ significantly. The expression of TNF-α in the renal tissue of SHR was higher than those of Wisters. QYYY granules could reduce the expression of TNF-α significantly. Conclusion QYYY granules had a therapeutic and protective effect on hypertension and early renal damage of hy ension to some extent. It should be related to lower content of AngⅡ and exp
