HPLC测定湘莲莲子心中莲心碱的含量

摘　　要：目的:采用HPLC测定了6个不同产地的湘莲莲子心中莲心碱的含量。方法:色谱柱为ODS C18(250mm×4.6mm,5μm);流动相为乙腈-0.015mol·L-1十二烷基磺酸钠-冰醋酸(56∶43∶1);体积流量为1mL·min-1;检测波长为282nm。结果:莲心碱在质量浓度0.0110～0.1210mg·mL-1与峰面积呈良好的线性关系,平均回收率99.14%,RSD=1.85%。湖南6个不同产地的湘莲莲子心生物碱的含量均在0.21%～0.32%。结论:测定的6个产地湘莲莲子心中莲心碱的含量均大于0.2%,符合《中国药典》2010版规定的要求,以产自湘潭的莲子心中莲心碱的含量最高,为0.32%。Objective：To determine Liensinine in Nelumbinis Plumula from six different areas of Hunan province by HPLC method.Methods：The ODS C18（250 mm×4.6 mm,5 μm）column was used with the mobile phase of acetonitrile-0.015 mol·L^-1 Sodium dodecylsulphate-acetic acid（56∶43∶1）at detection wavelength of 282 nm.The flow rate is 1 mL·min^-1.Results：The linear ranges of liensinine was 0.011 0 - 0.121 0 mg·mL^-1.The average recovery was 99.65%,and RSD was 0.76%.The Liensinine content in Nelumbinis Plumula from six different areas were 0.21%-0.32%.Conclusion：The Liensinine content in Nelumbinis Plumula from six different areas were all 〉 0.2%,according with the regulation of Chinese Pharmacopoeia which was published in 2010.The liensinine in Nelumbinis Plumula from Xiangtan was 0.32% and it was the highest in the six samples.
