摘 要:[目的]为高山雪鸡和藏雪鸡的选育、保护和利用提供科学依据。[方法]应用聚丙烯酰胺凝胶电泳法对10只高山雪鸡和10只藏雪鸡血清酯酶同工酶多态性进行研究,与20只家鸡进行了比较。[结果]高山雪鸡、藏雪鸡和家鸡血清酯酶均存在ES1、ES2和ES3 3种同工酶,且ES3均为单态。高山雪鸡ES1同工酶以ES1 ABC为优势表型(70.00%),藏雪鸡和家鸡ES1同工酶均以ES1B为优势表型(80%和75%)。高山雪鸡ES2同工酶只有1条浓染的酶活性区带。藏雪鸡ES2同工酶则表现为有带(+)和无带(-)2种区带,而家鸡ES2同工酶无显现酶活性区带。[结论]高山雪鸡和藏雪鸡ES同工酶多态性存在较大差异,也表明ES同工酶的区带数、着染强度、迁移率、基因表型等可作为区分禽类亲缘关系的生化遗传标记。[Objective] The research aimed to provide scientific basis for the breeding,protection and utilization of Tetraogllus himalayensis and Tetraogallus tibetanus.[Method] Using polyacrylamide gel electrophoresis,the isozyme polymorphism of serum esterase in 10 Tetraogllus himalayensis and 10 Tetraogallus tibetanus were studied and they were compared with that in 20 chicken.[Result] There were three isozymes(ES1,ES2 and ES3) in T.himalayensis,T.tibetanus and chicken and ES3 showed a single state.The dominant phenotype of ES1 isozyme in T.himalayensis was ES1 ABC,which took 70.00%.The dominant phenotype of ES1 isozyme in T.tibetanus and chicken was ES1B,which took 80% and 75% respectively.ES2 isozymes in T.himalayensis only had a thick enzyme activity zone.ES2 isozyme in T.tibetanus was represented two zones of(+) and(-).But ES2 isozyme in chicken had no enzyme activity zone.[Conclusion] The isozyme polymorphism of Es had great differences between T.himalayensis and T.tibetanus.And it was also indicated that the zone number,staining degree,transfer rate,gene phenotype of ES isozymes could be used as the bio-chemical genetic markers for distinguishing the genetic relationship of birds.
