摘 要:以150个不同生态类型的大豆品种(系)为试材,分别用4种腐霉菌进行接种鉴定,筛选抗病性,并用256个SSR(simple sequence repeat)标记对不同抗性类型大豆品种进行基因组扫描,分析群体结构和连锁不平衡,同时用TASSEL软件中的GLM方法对大豆4种腐霉菌的抗性进行关联分析,T测验检测差异显著位点,进而发掘各关联位点的优异等位基因。结果表明:①参试大豆品种与256个SSR位点存在广泛的连锁不平衡,而且共线位点D'衰退较快,适合于连锁不平衡分析。SSR数据结构和亲缘分析表明参试品种分为7个亚群。②用关联分析方法得到与4种腐霉菌抗性关联的引物25个,T测验和抗性验证找到差异显著的关联等位变异位点12个,它们在参试品种中都有较大的效应值。③关联分析鉴定差异显著的等位变异位点与实际品种抗病性结果高度吻合。其关联分析中可信度高的等位变异位点和T测验中显著的等位变异位点相吻合。即与P.aphanidermatum抗性关联的位点为Satt191-1、Satt584-1和Satt584-2;与P.irregulare抗性关联的位点为Satt602-1和Satt042-4。By using 150 soybean varieties with different ecotype as materials,and 4 Pythium species as inoculation pathogens, the cuhivars resistance were identified through measuring the root rot level. 256 SSR markers were used to analysis the popula- tion structure and linkage disequilibrium. A soybean resistance association analysis to 4 Pythium species was run with GLM procedure from software TASSEL, and a T test on associated markers with significant difference was carried on, to uncover asso- ciated loci conferring effects. The result showed:there was a comprehensive linkage disequilibrium;the collinear locus D'de- clined quickly, which made it fit for LD analysis ; tested cultivars were divided into seven subgroups by the structural analysis of SSR data. There were 25 markers obtained by association analysis,which were connected with the resistance to 4 Pythium spe- cies, respectively;12 association loci with significant difference,were found confering major effect values to the tested cultivars through T test and resistance identifying. The loci with significant difference highly matched the cultivars' resistant testing re- suits ; especially the ones with high reliability in association analysis showed the same on T test, such as the loci associated with P. aphanidermatum resistance, Satt191-1, Satt584-1, Satt584-2, and those with P. irregular resistance, Satt602-1, Satt042-4.
