摘  要:目的观察放疗后腮腺损伤过程中凋亡相关分子半胱氨酸天冬氨酸蛋白酶3(Caspase-3)、bcl-2相关x蛋白(Bax)和B细胞淋巴瘤/白血病-2(bcl-2)、P53在腮腺的时空表达,探讨肿节风在放射引起的腮腺损伤中的防护作用。方法将猪龄2~3个月的60头小型猪随机分成空白对照组(20只小型猪)、单纯照射组(20只小型猪)和肿节风加照射组(20只小型猪)3组,空白对照组(空白组)给予0Gy照射,单纯照射组(单照组)及肿节风加照射组(药照组)在全麻状态下给予总量为30Gy 60 COγ射线双侧腮腺照射,30Gy/(5f.5w)建立放射损伤模型。且药照组于照射前1周开始给予肿节风颗粒,直至腮腺组织取出,空白组与单照组给予等量生理盐水。每组根据不同处死时间再分为a、b、c、d四个亚组,分别于照射结束后1、10、40、90天处死后取腮腺组织。采用RT-PCR法检测各组腮腺细胞中Bcl-2、Bax、P53和Caspase-3mRNA表达情况。结果在同一时点(同一平行组),bcl-2mRNA表达量为空白组>药照组>单照组;bax、P53、Caspase-3mRNA表达量均为单照组>药照组>空白组;bcl-2mRNA表达量在照射结束后10天最低、90天最高;bax蛋白在10天最高、90天最低;药照组中,bcl-2mRNA表达量逐渐递增;Bax mRNA表达量逐渐递减(P<0.05)。单照组中P53和Caspase-3mRNA的表达量均呈逐渐增高趋势,药照组中,P53及Caspase-3mRNA的表达量也呈现逐步升高趋势,但各时间段表达量均低于单照组(P<0.05)。结论肿节风抑制腮腺细胞放疗后细胞凋亡的作用,可能是通过抑制Bax、P53和Caspase-3mRNA的表达,同时上调bcl-2mRNA的表达来实现。Objective To observe the expression of apoptosis--related molecules caspase--3, bcl--2 relat ed x protein (bax), B cell lymphoma/leukemia--2 (bcl--2) and P53 in radiation--injured parotid glands at different time and space points, and to explore the effect of Glabrous Sarcandra Herb on protection of parotid glands radiation injury. Methods 60 miniature pigs aged two months were randomly divided into 3 groups. normal control group (20 miniature pigs), radiation control group (20 miniature pigs) and medicine radiation group (20 miniature pigs). The normal control group received 0 Gy gamma--rays, the radiation control group and the medicine radiation group received a single dose of 30 Gy 60CO gamma--rays delivered to each parotid gland, the radiation injured models had been completed with 30 Gy/(5f · 5w). Glabrous Sarcandra Herb were administered to the medicine radiation group a week before exposure to the radiation, and equal physiological saline for the others. According to different death time, each group was divided into four subgroups a,b,c and d. Bilateral parotid glands tissues were taken out on days 1, 10, 40 and 90 following the exposure in subgroups a, b,c and d, respectively. The expression of Bcl--2,Bax,P53 and Caspase3 mRNA in parotid glands cells was detected by RT--PCR. Results At the same time (parallel to the same group), the expression of bcl--2 mRNA in each group was ranked: normal control group〉medicine radiation group〉radiation control group. All the expressions of bax, P53, Caspase--3 mRNA in each group were ranked radiation control group〉medicine radiation group 〉 normal control group. The expression of bcl--2 mRNA was the lowest on day 10 and highest on day 90. The expression of bax protein was the highest on day 10 and the lowest on day 90. In medicine radiation group, the expression of bcl--2 mRNA increased gradually, while the expression of bax mRNA decreased gradually ( P 〈0.05).
