摘  要:目的检测VEGF-C及COX-2在早期宫颈癌组织中的表达,探讨二者之间的关系及其在宫颈癌发生发展和淋巴结转移中的作用。方法收集武汉中心医院2005~2009年手术切除的宫颈癌组织标本57例,应用VEGF-C和COX-2多克隆抗体进行SP法免疫组化染色。用COX-2特异性抑制剂NS398处理宫颈癌HeLa细胞,应用Western blot方法检测HeLa细胞中VEGF-C蛋白的表达变化。结果 57例宫颈癌标本中,VEGF-C表达阴性(-)7例(12.28%),弱阳性(+)14例(24.56%),中等阳性()20例(35.09%),强阳性()16例(28.07%)。随着病变由宫颈原位癌到早期浸润癌的进展,VEGF-C表达明显增强。COX-2表达阴性(-)38例(66.67%),阳性(+)19例(33.33%)。VEGF-C和COX-2阳性染色均位于肿瘤细胞的胞质,在COX-2表达阳性部位,VEGF-C大多数呈强阳性反应,且二者均在癌巢中强表达。统计分析表明VEGF-C和COX-2表达呈正相关(P<0.01)。26例伴淋巴结转移者中,VEGF-C表达阳性21例,统计分析显示VEGF-C表达与淋巴结转移密切相关(P<0.05)。当用COX-2特异性抑制剂NS398处理HeLa细胞后,细胞中VEGF-C蛋白表达较处理前明显降低。结论 VEGF-C可能在宫颈癌癌变过程中起一定的促进作用,VEGF-C表达与宫颈癌细胞的淋巴结转移密切相关,检测活检标本VEGF-C表达情况可能有助于预测盆腔淋巴结转移。在宫颈癌组织中,VEGF-C和COX-2表达之间具有明显的相关性,COX-2可能是宫颈癌细胞中VEGF-C表达的一个调节者。Objective To investigate the expression of VEGF-C and cyclooxygenase-2(COX-2)in human cervical cancer at early stage,their correlation and their roles in tumor progression and lymph node metastasis of cervical cancer.Methods A total of 57 samples were collected from cervical cancer patients who underwent operation in Wuhan Central Hospital between 2005 and 2009.The expression of VEGF-C and COX-2 was immunohistochemically detected in the tissue samples.HeLa cells were cultured and treated with the COX-2-specific inhibitor NS398.Western blot was employed to detect the expression of VEGF-C protein.Results Both VEGF-C and COX-2were found to express in the cytoplasm of cervical cancer cells.In the 57 samples of cervical cancer,there were 7cases(12.28%)negative for VEGF-C,14(24.56%)weakly positive for VEGF-C,20(35.09%) moderately positive for VEGF-C and 16(28.07%)intensely positive for VEGF-C.The expression of VEGF-C was increased with the progression of cervical cancer.There were 38 cases(66.67%)negative for COX-2and 19(33.33%)positive for COX2.VEGF-C was strongly expressed in most COX-2-postive sites and they both were intensely expressed in cancer nests.The statistical analysis revealed that VEGF-C expression was positively correlated with COX-2expression(P〈0.01).In 26 cases of cervical cancer with lymph node metastases,VEGF-C was expressed in 21 cases and the VEGF-C expression was closely related to lymph node metastases(P〈0.05).The expression of VEGF-C was significantly decreased in HeLa cells treated with COX2-specific inhibitor NS398.Conclusion Given these observations,we conclude that VEGF-C and COX-2 may contribute to the occurrence and progression of cervical cancer;VEGF-C expression is closely associated with lymph node metastases in the pelvic cavity;there is a close relationship between the expression of VEGF-C and COX-2;COX-2 may be a regulator of VEGF-C expression in human cervical carcinoma cells.
