摘  要:目的探讨大鼠卵巢自初始发育至衰老过程中卵巢颗粒细胞及其细胞因子的变化规律及补肾调肝方药干预影响,以期为补肾调肝方药抗卵巢衰老的作用机制研究提供理论依据。方法分别选取3月、6月、12月、18月、24月龄Wistar大鼠模拟其青春前期、青春期、性成熟期、近绝经期、老年期的生长发育过程;各月龄大鼠均设立干预组和对照组,干预组以补肾调肝方灌胃干预20ml/(kg·d),对照组灌以同等剂量的无菌饮用水。以常规TUNEL染色法观察各组大鼠卵巢颗粒细胞状态,计算凋亡指数(AI);并以RT-PCR对颗粒细胞促凋亡因子Bax及凋亡抑制因子Bcl-2的mRNA表达进行半定量分析。通过各月龄组之间的对比观察卵巢颗粒细胞及其细胞因子的变化规律,并通过干预组和对照组之间的对比对补肾调肝方药抗卵巢衰老的相关机制做相应探讨。结果 AI:对照组3、6、12月龄大鼠的AI均处于较低,18、24月龄大鼠AI均显著升高(P<0.05);与对照组相比,研究组大鼠各月龄AI均显著降低(P<0.05)。Bax:对照组3、6、12月龄大鼠的BaxmRNA表达处于较低水平,18、24月龄显著升高(P<0.05);与对照组相比,干预组各月龄大鼠BaxmRNA表达均显著降低(P<0.05)。Bcl-2:对照组3、6、12月龄大鼠Bcl-2mRNA表达处于较高水平,18、24月龄显著降低(P<0.05);与对照组相比,干预组各月龄大鼠Bcl-2mRNA表达均显著升高(P<0.05)。结论凋亡抑制因子Bcl-2mRNA低表达及促凋亡因子BaxmRNA高表达所启动的颗粒细胞调亡过程可能为卵巢衰老的重要机制之一;补肾调肝方药可通过上调Bcl-2mRNA表达与下调BaxmRNA表达抑制卵巢颗粒细胞的调亡,可能为其抗卵巢衰老的作用机制之一。Objective To investigate the effects of rat ovarian changes from the initial development to the aging process in ovarian granulosa cells and their cytokines and Bushen tiaogan prescription intervention, and provide a theoretical basis for mechanism of Liver-regulating compound recipe for invigorating the kidney against ovarian aging in order to.Methods Simulated the prepubertal, pubertal, sexual maturity, close menopause, senile growth process in 3,6,12,18,24 months old wistar rats; each month old rats were established in the intervention group and the control group, the intervention group with Bushen tiaogan Fang gavage interference 20 ml/(kg·d), the control group was treated with the same dose of sterile drinking water.To observe the state of each rat ovarian granulosa cells with routine TUNEL staining, apoptotic index (AI); a semiquantitative analysis of the expression and RT-PCR on granulosa cells apoptosis and promote Bax apoptosis inhibition factor Bcl-2 mRNA.Changes of ovarian granulosa cells and their cytokines by comparison between the ages of, corresponding to explore the relevant mechanisms and through comparison between the intervention group and the control group of Bushen tiaogan recipe against ovarian aging.Results AI: control group 3, 6, 12 months age rats AI are low, 18, 24 month old rats AI increased significantly (P0.05); compared with the control group, the study group of rats at different ages AI were significantly decreased (P0.05).Bax: control group, 6 December 3, aged rat Bax mRNA expression at low levels, 18, 24 months of age were significantly increased (P0.05); compared with the control group, the intervention of mRNA Bax group each month old rats were significantly decreased (P0.05).Bcl-2: control group 3, 6, December age rat Bcl-2 mRNA expression in a high level, 18, 24 months of age, decreased significantly (P0.05); compared with the control group, the intervention of mRNA Bcl-2 group each month old rats were significantly increased (P0.05).Conc
