益气活血颗粒对大鼠心气虚证cAMP/cGMP影响的研究

摘　　要：目的:观察益气活血颗粒对大鼠心气虚证cAMP/cGMP的影响。方法:用大鼠负重强迫游泳后通过给予大剂量心得安使大鼠造模定位于心脏,作为心气虚证实验动物模型。分别以益气活血颗粒、天王补心丸、生理盐水灌胃。于大鼠胸壁心跳最明显处,将针头刺入心脏,分别取出2～3mL血液。观察大鼠血浆和心肌中cAMP、cGMP及cAMP/cGMP的变化。结果:造模组与各组间比较均有显著性差异(P<0.01、P<0.05),大剂量治疗组分别与小剂量、阳性对照组比较有显著性差异(P<0.01)。心肌中小剂量治疗组与阳性对照组比较有显著性差异(P<0.01)。结论:心气虚证模型成立。益气活血颗粒可升高cAMP含量,降低cGMP含量。Objective To observe the influence of some Chinese herbal granules with the effects of replenishing qi and activating blood on the cAMP/cGMP in big-rats with syndrome of heart-qi insufficiency.Methods：The model was established by big-rats freeweight compeling swimming and afforded big dosing Propranolol location on heart,and given orally Chinese herbal granules,Tianwang buxin Pills and normal saline,respectively.Locate big-rats evident heartbeat,with pinhead penetrating into heart so that took out 2-3ml blood.Observe big-rats cAMP,cGMP,cAMP/cGMP in the blood plasma and cardiac muscle.Results：The comparison was significantly different between model group and other different groups（P〈0.01,P0.05）.The comparison was significantly different between the high-dose group（P〈0.01） and the little-dose group.The comparison was significantly different between the high-dose group and positive control group（P〈0.01）.The comparison was significantly different between the small-dose group and positive control group in the cardiac muscle（P〈0.01）.Conclusion：The model with the syndrome of heart-qi insufficiency is tenable.The Chinese herbal granules with the effects of replenishing qi and activating blood are effective for raising cAMP content and reducing cGMP content.
