摘 要:通过PCR-单链构象多态性(PCR-SSCP)结合测序技术发现FoxO1第4外显子一个新单核苷酸多态性(SNP),限制性片段长度多态性聚合酶链反应(PCR-RFLP)技术分别分析925头中国荷斯坦奶牛FoxO1该位点多态性。结果表明,在中国荷斯坦奶牛中,FoxO1基因有3个基因型,分别是CC、TT、CT。其中等位基因C频率为0.95,是优势等位基因;χ2适合性检验表明该位点不符合哈代温伯格平衡(P<0.05)。该基因不同基因型对奶牛产犊间隔、空怀期以及初产日龄有显著影响(P<0.05),而对产犊到发情、犊牛初生重、配种次数影响不显著(P>0.05)。PCR-single-strand conformation polymorphism (PCR-SSCP) and sequencing are used and a novel single nucleotide polymorphisms (SNP)was detected in the forth exon of FoxO1 gene. The gene types of the 925 Holsteins were differentiated by PCR-restriction fragment length polymorphism (PCR-RFLP). The result showed that there were three genotypes (CC, CT, TT) and the frequency of allele C which was superior gene was 0.95; χ2 fitness test showed that the loci deviates from the Hardy-Weinberg equilibrium (P〈0.05). The correlation analysis revealed that these polymorphisms had a extremely significant effects on calving interval, open days and the first calving age (P〈0.05), but there was not a significant difference with the days between calving and oestrus, the times of service per conception and birth weight (P〉0.05).
