In vivo and liver microsome in vitro analysis of metabolites of enrofloxa cin from crucian carp (Carassius auratus Gibelio) using UPLC/Q-TOF MS

摘  要:采用超高效液相色谱-四级杆-飞行时间质谱法(UPLC/Q-TOF MS)对恩诺沙星在异育银鲫体内和体外肝微粒体中代谢产物进行定性分析。结果表明,异育银鲫血浆中除以恩诺沙星原形药物为主外,还可检测到少量N-去乙基代谢产物环丙沙星、葡萄糖醛酸结合物;而在肝微粒体体外孵育体系中除检测到原形药物外,只有少量N-去乙基代谢物环丙沙星。无论是体内还是体外,代谢产物生成量都很少。推测恩诺沙星在异育银鲫体内代谢反应主要是脱乙基反应和葡萄糖醛酸结合反应,而在体外肝微粒体恩诺沙星代谢反应主要是脱乙基反应。An ultraperformance liquid chromatography coupled with Q-TOF mass spectrometry (UPLC/ Q-TOF MS) was used for the identification of metabolites of enrofloaxcin in crucian carp (Carassius auratus Gibelio) in vivo and in liver microsome in vitro. Two metabolites, ciprofloxacin and enrofloxacin glucuronide, were detected in crucian carp plasma after oral administration of enrofloxacin. However, in the crucian carp liver microsome incubation in vitro, only one metabolite of ciprofloxacin was found. Whether in vivo or in vitro, very little amount of metabolites were biotransformed. It was speculated that enrofloxacin metabolism in crucian carp in vivo might be de-ethylation and glucuronidation, while in liver microsome in vitro being de-ethylation.
