摘  要:目的建立液相色谱-质谱(LC-MS)测定人血浆中氢氯噻嗪浓度的方法。方法以对乙酰氨基酚为内标,血浆样品经乙酸乙酯提取后进样测定。分析柱为XTerraMS C18柱(5μm,2.1 mm×150 mm);保护柱为Diamonsil C18(2)Gart-ridges(3μm,2.1 mm×10 mm),柱温30℃;流动相为水:乙腈-60:40,流速为0.2 ml/min。用于定量的检测离子为氢氯噻嗪296,内标150。结果血浆氢氯噻嗪浓度在0.5~150μg/L范围内线性关系良好,方法回收率为95.6%~109.1%,日内、日间RSD均小于15%。结论所建立的LC-MS方法灵敏、准确、快速,可用于人血浆中氢氯噻嗪浓度的测定。Objective To establish a LC-MS method for determination of hydrochlorothiazide in human plasma. Methods Acetaminophen was used as internal standard (IS). After extraction with ethyl acetate, the concentrations of hydrochlorothiazide in plasma were measured by LC-MS. The determination was performed on XTerra MS C18(2)Gartridges(3μm,2.1mm×10mm) column and the col- umn temperature was 30℃. The mobile phase was H20-acetonitrile (60 : 40) and the flow rate was 0. 2 ml/min. Target ions were m/ z 296 for Hydrochlorothiazide and m/z 150 for acetaminophen (IS) ,respectively. Results Calibration curve of the method was linear within the range of 0.5~150μg/L. The method recovery was 95.6% - 109. 1%. RSDs of intra-day and inter-day were all less than 15%. Conclusions The method was sensitive accurate and rapid that could be used for determining the concentration of hydrochlo- rothiazide in human plasma.
