摘 要:目的探讨姜黄素对大鼠肾缺血再灌注损伤后核因子-κB(NF-κB)表达的影响。方法 24只大鼠随机分为3组:假手术组、缺血再灌注组和姜黄素组,每组8只,在肾组织缺血45 min后完全恢复组织灌流。全自动生化分析仪检测肌酐(Cr)、尿素氮(BUN),光镜下观察肾组织病理学改变,Western blotting法检测NF-κB蛋白表达。结果与假手术组比较,缺血再灌注组的肾功能指标水平明显增高,光镜下可见肾小管损伤明显加重,Western blotting显示NF-κB表达明显增高,差异有统计学意义(P<0.05)。姜黄素组与缺血再灌注组比较,肾功能指标明显减轻,NF-κB表达显著降低,差异有统计学意义(P<0.05)。结论姜黄素能减轻肾缺血再灌注损伤,其机制可能是抑制NF-κB的表达有关。Objective To investigate the effects of curcumin on expression of nuclear factor-κB (NF-κB) in renal ischemia and reperfusion injury. Methods 24 rats were randomly divided into 3 groups: sham group, ischemia and reperfusion injury group, and curcumin group. Each group had 8 rats. The rats were made complete reperfusion after 45 min renal ischemia. The parameters (Cr, BUN) of renal function were determined by auto-biochemical analyzer.And then renal histopathological changes were observed and NF-κB protein expression were detected by Western blotting. Results Compared with sham group, Cr and BUN levels in the group of ischemia and reperfusion increased significantly, and histopathological damage of renal tubular aggravated. Western blotting showed that NF-κB expression increased significantly, difference was statistically significant (P 〈 0.05). Compared with ischemia and reperfusion group, the damage of curcumin group significantly reduced (P 〈 0.05). Conclusion Curcumin can ameliorate renal ischemia-reperfusion injury and its mechanism may be through inhibiting the expression of NF-κB.
