胆囊胆固醇结石仓鼠胆囊平滑肌及Oddi括约肌中Caveolins、P450arom及黏蛋白的表达研究

摘　　要：目的探讨胆囊胆固醇结石仓鼠胆囊平滑肌及Oddi括约肌中Caveolins、P450arom及黏蛋白的表达情况。方法将100只仓鼠随机分为A组(常规饮食组)20只、B组(致石饮食2周组)20只、C组(致石饮食4周组)20只、D组(致石饮食6周组)20只和E组(致石饮食8周组)20只,检测5组仓鼠的胆囊平滑肌及Oddi括约肌Caveolin1、Caveolin3、P450arom及黏蛋白1、黏蛋白2、黏蛋白3的阳性表达率并进行比较。结果 E组胆囊平滑肌及Oddi括约肌中Caveolin1、Caveolin3、P450arom及黏蛋白1、黏蛋白2的阳性表达率均高于其他4组,D组高于A、B、C组,C组高于A、B组,差异均有统计学意义(P<0.05)。E组胆囊平滑肌及Oddi括约肌中黏蛋白3阳性表达率低于其他4组,D组低于A、B、C组,C组低于A、B组,差异均有统计学意义(P<0.05)。上述指标均与胆囊胆固醇结石有明显的相关性(均P<0.05)。结论胆囊胆固醇结石仓鼠胆囊平滑肌及Oddi括约肌中Caveolins、P450arom及黏蛋白的表达呈现异常,对了解胆囊胆固醇结石的形成有一定意义。Objective To study the expression situation of Caveolins, P450arom and mucoprotein in gallbladder smooth muscle and Oddi sphincter of hamsters with cholesterol gallstone. Methods 100 hamsters were randomly divided into group A （routine diet group） of 20 cases, group B （2 weeks of lithogenic diet） of 20 cases, group C （4 weeks for lithogenic diet） of 20 cases, group D （6 weeks of lithogenic diet） of 20 cases and group E （8 weeks of lithogenic diet） of 20 cases. Then the positive expression rates of Caveolinl, Caveolin3, P450arom and mucoprotein 1, mucoprotein 2, mucoprotein 3 of the 5 groups were detected and compared. Results The positive expression rates of Caveolinl, Caveolin3, P450arom and mucoprotein 1, mucoprotein 2 in gallbladder smooth muscle and Oddi sphincter of group E were all higher than those of other 4 groups, those of group D were higher than group A, B, C, those of group C were higher than group A and group B, the differences were statistically significant （P 〈 0.05）. While the positive expression rates of mucoprotein 3 in gallbladder smooth muscle and Oddi sphincter of group E were lower than those of other 4 groups, those of group D were lower than group A, B, C, those of group C were lower than group A and group B, the differences were statistically significant （P 〈 0.05）. Those indexes all had obvious correlation to cholesterol gallstone （all P 〈 0.05）. Conclusion The expression of Caveolins, P450arom and mucoprotein in gallbladder smooth muscle and Oddi sphincter of hamster with cholesterol gallstone are abnormal,and it has some significance for understanding the formation of cholesterol calculus.
