摘 要:目的采用响应面法对金色链霉菌发酵产金霉素培养基进行优化。方法首先采用Plackett-Burman实验筛选出影响金色链霉菌发酵产生金霉素的主要影响因素,然后通过最速上升实验逼近最大值响应区域,最后采用Box-Behnken设计响应面方法确定主要影响因素的浓度,得到金霉素优化发酵培养基组成。结果玉米淀粉、酵母粉、(NH4)2SO4为对金霉素产量有显著影响的主要因素,最佳水平为:玉米淀粉125.10 g.L-1、酵母粉4.03 g.L-1、(NH4)2SO44.87 g.L-1。在优化发酵培养基条件下,金霉素发酵单位为22.813 g.L-1,与预测值接近,较优化前发酵单位19.476 g.L-1提高了17%。结论响应面实验设计和分析方法能够有效地对金霉素发酵培养基进行优化。Objective To optimize the fermentation medium of Streptomyces aureofacien for chlortetracycline production by response surface methodology.Methods First,the Plackett-Burman design was applied to screen out the major factors influencing the production of chlortetracycline.Subsequently,the steepest ascent experiments were performed to approach the optimal region of the medium composition.Finally the optimal concentrations of the major factors were determined and the optimized fermentation medium was obtained through response surface methodology of Box-Behnken design.Results Corn starch concentration,yeast powder concentration and(NH4)2SO4 concentration were found to be the major factors affecting chlortetracycline production significantly.And their optimal levels were observed when 125.10 g · L-1 of corn starch,4.03 g · L-1of yeast powder and 4.87 g · L-1of(NH4)2SO4 were used.Under the optimized medium,the yield of chlortetracycline was 22.813 g · L-1,which was close to the predictive value.The production was increased by 17% compared with that under the original medium,which was 19.476 g · L-1.Conclusions The experimental design and analysis of response surface methodology can be used to optimize effectively the fermentation medium for chlortetracycline production.
