摘 要:目的:研究雷公藤内酯对癫痫模型大鼠海马神经元凋亡的保护作用。方法:皮下注射海人酸(10mg/kg)以复制大鼠癫痫模型。实验分为对照(等容生理盐水)、模型(等容生理盐水)与雷公藤内酯(15μg/kg)组,腹腔注射给药,每天1次,连续7d。结晶紫染色观察海马神经元形态;免疫组化染色和Western blot法检测海马神经元B细胞淋巴瘤/白血病-2(Bcl-2)、Bcl-2相关X蛋白(Bax)表达水平。结果:模型组大鼠海马神经元变性或丢失,胞体皱缩,形态不规则;雷公藤内酯组大鼠海马神经元的数量和形态明显改善。与对照组比较,模型组大鼠Bcl-2蛋白表达水平显著减弱,Bax蛋白表达显著增强(P<0.05);与模型组比较,雷公藤内酯组大鼠Bcl-2蛋白表达水平显著增强,Bax蛋白表达显著减弱(P<0.05)。结论:雷公藤内酯可通过上调癫痫模型大鼠海马神经元Bcl-2、下调Bax蛋白表达而抑制大鼠神经元凋亡。OBJECTIVE: To investigate the protective effects of triptolide on neuronal apoptosis in detect the epileptic model rats. METHODS: Epileptic model was induced by hypodermic injection of kainic acid (10 mg/kg). Model rats were divided into control group (constant volume of normal saline), model group (constant volume of normal saline) and triptolide group (15 μg/kg). They were given relevant medicine intraperitoneally once a day for consecutive 7 days. Crystal violet staining was used to observe the morphological change of the hippocampal neuron. immunohistochemistry staining and Western blot assay were used to examine the expression of Bcl-2 and Bax protein in the hippocampal neuron. RESULTS: Crystal violet staining showed that the neurons of the hippocampal region exhibited neuronal degeneration or loss, body shrinkage and irregular morphology in model group. The number and morphology of neurons in triptolide group had been improved significantly. Compared with control group, the expression of Bcl-2 protein in model group was decreased significantly, while the expression of Bax protein was increased significantly(P〈0.05). Compared with model group, the expression of Bcl-2 in triptolide group was obviously increased, while the expression of Bax was obviously decreased(P〈0.05). CONCLUSIONS: Triptolide can inhibit the neuronal apoptosis of rats through up-regulating the expression of down-regulating Bcl-2 and Bax protein in the hippocampal region.
