摘  要:研究利用简易褪色光度法测定马蹄皮提取物清除羟自由基能力的新体系。在表面活性剂存在下的酸性介质中,利用Fenton反应产生的羟自由基能迅速氧化吖啶红-罗丹明混合染料体系使其褪色,利用马蹄皮提取物对羟自由基的清除作用,使混合染料褪色程度降低,据此建立测定抗氧化能力的新方法,探讨了最佳的实验条件。方法测定羟自由基的相对标准偏差为2.82%,马蹄皮不同提取物的清除率次序为乙醇提取物>乙酸乙酯提取物>甲醇提取物>水提取物。该体系与单一体系相比,混合体系发生离子缔合,提高了方法的灵敏度和准确度,方法具有较好选择性和稳定性,可以应用于判断天然提取物的抗氧化能力。A new determination system for hydroxyl radical and antioxidant activity of tuberose peel extraction by simple fading spectrphtomtry.The hydroxyl radical produced by Fenton reaction can decrease the color intensity of Acridine Red-Rhodamine B alkaline dyestuff system. The change in absorbance is measured, and the amount of OH can thus be determined indirectly. The extractions of eleocharis tuberose peel could eliminate hydroxyl radi- cal in solution, hence its weakening effect of absorbance will be minimized. According to this principle, a new method for the determination of scavenging percentage of hydroxy radical with antioxidant was developed. The re- sults showed that the ability to scavenge hydroxyl radicals of various solvent extracts from Eleocharis tuberose peel was in the order of ethanol fraction〉ethyl acetate fraction〉methanol fraction〉water fraction. The sensitivity and ac- curacy of the mix system were raised compared to single system; the method has good selectivity and stability to apply in judging oxidation resistance of natural extraction.
