摘 要:试验采用稀释平板法分离瘤胃细菌,通过刚果红平板法初筛和大豆秸秆遥瓶复筛,从奶牛瘤胃内容物中分离筛选一株能高效分解大豆秸秆粗纤维的菌株。结果表明,该菌株的分解能力强,可使大豆秸秆中NDF从86.86%下降至77.43%,ADF从58.60%下降至46.15%。对该菌株进行初步鉴定和培养条件优化结果表明,该菌株为革兰氏阴性无芽孢杆菌,培养时间37℃、培养温度60 h、接种量10%、氮源添加量4%为该菌最适产酶条件。In order to screen out bacteria that could decomposes soybean straw crude fiber efficiently from rumen flu- id,the methods adopted that trial separation of rumen bacteria by the dilution plate and Congo red medium plate method, and soybean straw flask shaking fermentation method.The results showed that:the strain could decompose soy bean straw, and it could make the NDF of soybean straw decrease from 86.86% to 77.43% and the ADF of soybean straw decrease from 58.60% to 46.15%.Thongh the preliminary identification of strains and optimization of culture medium,it was Gram- negative bacillus.The optimum conditions for enzyme production were culture temperature 37 ℃,culture time 60 h,inocu- lation-quantity 10%, nitrogen source adding quantity 4%.
