摘 要:目的建立双标定量指纹法精确恢复银翘解毒丸对照指纹图谱(RFP)标准,以实现动态指纹图谱标准的校正和复原。方法用RP-HPLC法对银翘解毒丸(YQJDP)中绿原酸、咖啡酸、连翘苷和牛蒡苷4组分进行定量分析。建立228、265和280 nm下YQJDP-HPLC指纹图谱,采用均值法整合三波长下定性定量全信息,应用六级系统指纹定量法对其进行评价。选择绿原酸和牛蒡苷为双参照物峰(双标),采用双标定量指纹法恢复YQJDP-RFP。结果三波长整合指纹图谱评价结果如下:S3、S4、S6和S8质量为极好(1级),S2、S5、S7和S9质量为很好(2级),DS1质量中(5级),S1、DS3、DS4、WS1和WS2质量次(7级),DS2质量劣(8级)。含量测定结果与指纹图谱评价结果很一致。在此基础上建立YQJDP双标定量指纹法技术标准。结论基于多指标定量分析和多波长HPLC指纹谱的双标定量指纹法能够找到产生RFP的标准制剂和实现实时随行定量检测YQJDP指纹图谱。Objective To establish the dual reference fingerprints quantified method(DRFQ) to recover the reference fingerprint(RFP) of Yinqiao Jiedu pill(YQJDP),and to realize the correction and recovery of a dynamic fingerprint standard.Methods Four components(chlorogenic acid,caffic acid,forsythin and arctiin) of YQJDP were quantitatively analyzed by RP-HPLC.Meanwhile 3 wavelength fingerprints were established under 228,265 and 280 nm and were integrated by the average method,and the quality grades were evaluated by level-6 systematically qualified fingerprint method(L6-SQFM).DRFQ was applied to correct and recover a dynamic fingerprint standard of YQJDP,with the peak of CGA and AIT as the dual reference peaks.Results The evaluation results were as follows: the quality of S3,S4,S6 and S8 was the best(grade 1),that of S2,S5,S7 and S9 was better(grade 2),that of DS1 was moderate(grade 5),that of S1,DS3,DS4,WS1and WS2 was defective(grade 7) and that of DS2 was inferior(grade 8).The 4 compound determinations were consistent with the evaluation results of the 3 wavelength fingerprints.The dynamic technique criteria of YQJDP were established by DRFQ.Conclusion Based on multi-component quantitative analyses and parallel 3 wavelength high performance liquid chromatography fingerprints,the standard prescription that can build RFP will be obtained and the real-time accompanying quantitative detection of YQJDP fingerprint can be realized by DRFQ.
