摘  要:为了进一步研究激素对颗粒细胞分泌孕酮的影响,试验采用体外培养绵羊卵巢壁颗粒细胞后,经单独、联合添加2.5 IU/mL卵泡刺激素(FSH)、促黄体生成激素(LH)和1μg/mL雌二醇(E2),并于0,24,48,72,96小时时收集培养液,通过ELISA方法检测孕酮含量。结果表明:联合添加激素组在24小时时有促进孕酮分泌的作用(P<0.05),但随着时间的延长,其孕酮分泌总体呈下降趋势;单独添加FSH和LH均未见有效刺激孕酮分泌。To further study the effects of hormones on the secretion of progesterone by granulosa cells, the ovine ovarian granulosa cells were cultured in virto, and then were added alone and in combination with 2.5 IU/mL follicle -stimulating hormone(FSH) ,luteinizing hormone(LH) and 1μg/mL estradiol( E2 ), respectively; the culture mediums were collected at 0 h, 24 h, 48 h, 72 h, and 96 h, respectively. The enzyme -linked immunosorbent assay (ELISA) assay was used to measure the content of progesterone. The results showed that the group in combination with 2.5 IU/mL FSH, LH and 1μg/mL E2 had a role in promoting the secretion of progesterone compared with the control in 24 h ( P 〈 0.05 ), but the secretion of progesterone overall was in a downward trend with the extension of time. FSH and LH couldn' t effectively stimulate the secretion of progesterone when added alone.
