摘  要:为了克隆五指山小型猪DAZ相关蛋白2(DAZAP2)cDNA全长并进行生物信息学分析,试验以构建的五指山小型猪外周血白细胞cDNA文库为材料,采用菌落PCR方法克隆得到DAZAP2全长cDNA序列,并运用生物信息学软件对其核苷酸序列进行分析,预测其编码蛋白的理化性质及二级结构等。结果表明:五指山小型猪DAZAP2基因的核苷酸序列及其氨基酸序列与人、苏门达腊猩猩、斑马鱼、非洲爪蟾、牛、褐家鼠等动物具有很高的相似性。DAZAP2 cDNA全长943 bp,5'非翻译区长69 bp,3'非翻译区长367 bp,含有1个完整的开放阅读框,编码168个氨基酸。该蛋白的分子质量为17 311 ku,等电点为7.48。To clone full - length cDNA of DAZ associated protein 2 ( DAZAP2 ) gene of Wuzhishan miniature pig and perform the bioinformatics analysis, a full - length of cDNA sequence of DAZAP2 gene was obtained by colony PCR, using the cDNA library as a template, which was es- tablished from the peripheral blood leucocytes in Wuzhishan miniature pig. The bioinformatics software was used to analyze the gene sequence, for the prediction of physicochemical properties and secondary structure in the encoded protein. The results showed that it was found that the nu- cleotide sequence and amino acid sequence of DAZAP2 gene were highly homologous to Homo sapiens, Pongo abelii, Danio redo, Xenopus lae- vis, Bos Taurus, Rattus norveg/cus and other animals reported. The bioinformatics analysis showed that the total length of DAZAP2 cDNA sequence was 943 bp, and the lengths of 5 'UTR and 3'UTR were 69 bp and 367 bp, respectively. The cDNA contained an open reading frame encoding a protein of 168 amino acids. Its relative molecular weight was 17 311 ku, and the isoelectric point was 7.48.
