摘  要:目的研究顺铂(DDP)联合伊立替康(Irinotecan)注入综合疗法对HeLa细胞和HEK-293细胞的影响,以评估该模型作为检测方法的有效性,为中晚期宫颈癌的综合化疗提供优化线索。方法不同浓度DDP,Irinotecan,及双药联合作用于对数生长期的HeLa或HEK-293细胞,通过形态学观察、四甲基偶氮唑盐法(MTT)细胞活力测定,Ki67染色等检测药物对细胞的影响。结果自10μg/mL DDP对HeLa和HEK-293细胞生长均有抑制作用。自10μM Irinotecan对HeLa细胞生长抑制明显。10μg/mL DDP与低至2.5μMIrinotecan联合对HeLa细胞抑制作用显著强于同浓度单药。相同联合用药对HEK-293细胞的抑制作用明显较弱。结论 DDP与Irinotecan联合应用,不但提高了DDP综合疗法的效果,而且降低了化疗药物的使用剂量,此类联合治疗可望减轻患者的不良反应。相同联合方案对两种细胞的不同疗效提示了HeLa细胞作为宫颈癌体外模型的特异性。该研究可为宫颈癌综合疗法顺铂联合化疗方案的制定提供理论指导。[Objective] To evaluate the combination effect of cisplatin (DDP) with Irinotecan on Hela and HEK- 293 cell to examine the effectiveness of established models ,and to provide optimization of neoadjuvant chemothera- py in cervical cancer. [Methods] HeLa cell and HEK-293 cells were subjected to DDP, Irinotecan, or both of them in various dosages. The morphology, MMT colorimetry, and Ki67 immunocytochemistry of the cells were observed at different time-point. [Results] Suppressive effects were observed on HeLa and HEK-293 cells with DDP from 101xg/mL. Remarkable inhibitions on growth of HeLa cell were found with 101xM Irinotecan. Obvious synergic effects were found on HeLa cell when 10p^g/mL DDP and 2.51xM Irinotecan were applied. These applications of DDP and/or Irinotecan obtained much weaker inhibitions on HEK-293 cell. [Conclusion] The combination of DDP and Irinote- can could not only enhance the sensitivity of DDP to HeLa cells, but also decrease the dosage of DDP. Potentially it suggests a clinical application of DPP with Irinotecan have less adverse reactions. Furthermore, the significant differ- ence of the applications in the HeLa and HEK-293 cells indicates specificity of HeLa cell as an ideal in vitro model of cervical carninoma. The present study may provide clues for optimization of neoadjuvant chemotherapy with cis- platin in cervical carcinoma.
