摘 要:目的探讨二甲基甲酰胺(dimethylformamide,DMF)对正常心肌细胞(H9C2)的细胞毒性作用。方法采用CCK-8法检测不同浓度梯度(0 mmol/L,50 mmol/L,100 mmol/L,150 mmol/L,200 mmol/L,250 mmol/L,300 mmol/L)DMF作用心肌细胞24 h、48 h、72 h后的细胞毒性,并用Annexin V-FITC/PI流式细胞术检测不同时间(24 h、48 h、72 h)和浓度(24 h的染毒剂量为0 mmol/L,100 mmol/L,140 mmol/L,180 mmol/L,220 mmol/L,250 mmol/L;48 h的染毒剂量为0 mmol/L,100 mmol/L,140 mmol/L,180 mmol/L,200 mmol/L,220 mmol/L;72 h的染毒剂量为0 mmol/L,100 mmol/L,125 mmol/L,140 mmol/L,180 mmol/L,200 mmol/L)的细胞凋亡率。结果不同浓度DMF作用于心肌细胞24 h,48 h和72 h后产生了明显的细胞毒性。同一染毒时间组内,不同剂量组的细胞存活率差异有统计学意义(均有P<0.05);同一染毒剂量组内,各不同染毒时间组的细胞存活率差异均有统计学意义(均有P<0.05)。用CCK-8法检测DMF作用于心肌细胞的24 h IC50是250 mmol/L,48 h IC50是220 mmol/L,72 h IC50是200 mmol/L。细胞凋亡趋势与其对细胞的毒性作用结果一致。结论与阴性组比较,DMF对心肌细胞产生了细胞损伤,具有细胞毒性。Objective To discuss the cell toxicity of normal myocardial cells (H9c2) which exposed to Dimethyl- formamide (DMF). Methods The survival rates of myocardial cells which exposed to different doses of DMF were detec- ted by CCK-8 in 24 h, 48 h, 72 h. The method of Annexin V-FITC/PI flow cytometry was used to detected the cell apop- tosis rate. The concentration of DMF were different in different time. In 24 h, the concentration were 0 mmol/L, 100 mmol/L, 140 mmol/L, 180 mmol/L, 220 mmol/L, 250 mmol/L. In 48 h, the concentration were 0 mmol/L, 100 mmol/ L, 140 mmol/L, 180 mmol/L, 200 mmoL/L, 220 mmol/L. In 72 h, the concentration were 0 mmo]/L, 100 mmoL/L, 125 mmol/L, 140 mmol/L, 180 mmol/L, 200 mmol/L. Results The obvious cytotoxicity appeared when the myocardial cells was exposed to different concentrations of DMF in 24 h, 48 h,72 h. Within the same time in the infected group, the cell viability in different dose groups of DMF had significant differences( all P 〈 0. 05 ) , within the same virus dose group, the cell viability in differut time infected group had significant difference ( all P 〈 0. 05 ). The IC50 of H9c2 24 h, 48 h and 72 h after their exposure to DMF were 250 mmol/L, 220 mmol/L and 200 mmol/L, which were detected by method of CCK- 8. The cell apoptosis was the same as the trend of cell toxicity. Conclusions Compared with the negative group, the dif- ferent concentration of DMF can produce evident cell damage and cytotoxicity.
