摘  要:目的观察金水宝胶囊对慢性支气管炎大鼠内皮素-1(ET-1)和一氧化氮(NO)含量的影响,探讨金水宝胶囊的作用机制。方法将50只SD大鼠随机分为5组,即空白对照组、模型对照组、金水宝胶囊低剂量组、金水宝胶囊高剂量组和阳性药对照组,每组各10只。除空白对照组外,其余4组大鼠采用脂多糖气管内滴注法复制慢性支气管炎大鼠模型。金水宝胶囊低、高剂量组分别予0.03、0.06 g/mL的金水宝胶囊混悬液;阳性药对照组予0.09 g/mL的桂龙咳喘宁胶囊混悬液;空白对照组、模型对照组予0.9%氯化钠注射液10 mL/kg。连续灌胃给药21 d后测定大鼠血液、肺组织中ET-1和NO含量。结果模型对照组大鼠血浆、肺组织中ET-1含量均高于空白对照组(P<0.01),说明造模成功。金水宝胶囊高剂量组、阳性药对照组大鼠血浆中ET-1含量均高于模型对照组(P<0.01),金水宝胶囊低剂量组、金水宝胶囊高剂量组、阳性药对照组大鼠肺组织中ET-1含量均高于模型对照组(P<0.01)。金水宝胶囊高剂量组、阳性药对照组大鼠血浆、肺组织中ET-1含量均低于金水宝胶囊低剂量组(P<0.05,P<0.01)。金水宝胶囊高剂量组大鼠血浆中ET-1含量低于阳性药对照组(P<0.05)。模型对照组大鼠血清、肺组织中NO含量均低于空白对照组(P<0.01),说明造模成功。金水宝胶囊低剂量组、金水宝胶囊高剂量组、阳性药对照组血清中NO含量均高于模型对照组(P<0.01),金水宝胶囊高剂量组、阳性药对照组肺组织中NO含量均高于模型对照组(P<0.01)。金水宝胶囊高剂量组血清、肺组织中NO含量均高于金水宝胶囊低剂量组、阳性药对照组(P<0.05,P<0.01)。结论金水宝胶囊可通过有效抑制ET-1的产生,显著提高NO含量,恢复肺脏血管、支气管平滑肌正常舒缩状态,控制肺脏慢性炎症。Objective To observe the effect of Jinshubao capsule on endothelin - 1 ( ET - 1 ) and NO in rats with chronic bronchitis. Methods 50 SD rats were randomly divided into five groups, blank control group, model con- trol group,Jinshuibao low- dose group ,Jinshuibao high- dose group and positive drug group, 10 rats in each group. Chronic bronchitis model rats were established by Lipopolysaccharide( LPS ) intratracheal instillation. 0.03g/ml Jin- shuibao ,0.06 g/mL Jinshuibao and 0.09ml/kg Guilongkechuanning were given rats in Jinshuibao low - dose group, Jinshuibao high -dose group and positive drug group. Rats in blank control group and model control group received 0.9% sodium chloride solution. The content of ET - 1 and NO in blood and lung tissue were measured after 21 d in- ragastric administration of rats. Results The content of ET - 1 in blood and lung tissue in model control group issignificantly higher than that in blank control group for description successful modeling. The content of ET - 1 in rats plasma in Jinshuibao high - dose group and positive drug group was higher than that in model control group (P 〈0.01 ). The content of ET- 1 in rats lung tissue in Jinshuibao low- dose group,Jinshuibao high- dose group and positive drug group was higher than that in model control group(P 〈0. 01 ). The content of ET - 1 in rats plasma and lung tissue in Jinshuibao high - dose groupand positive drug group was lower than that in 3inshuibao low - dose group (P 〈 0.05, P 〈 0.01 ). The content of ET - 1 in rats plasma in Jinshuibao high - dose group and was lower than that in positive drug group( P 〈 0.05 ). The con- tent of NO in serum and lung tissue in model control group is significantly lower than that in blank control group for description successful modeling. The content of NO in rats serum in Jinshuibao low - dose group, Jinshuibao high - dose group and positive drug group was higher than that in model control group (P 〈 0.01 ).
