摘 要:作者报告用酚试剂法测定血浆中Vit C的实验研究。结果表明,酚试剂能和Vit C反应产生一蓝色化合物,该化合物在波长750nm处有最大吸收峰,在1.14-45.44μmol/L浓度范围内,吸收率和Vit C浓度呈线性关系(r=0.999,P<0.01)。Vit C标准品和酚试剂产生的颜色从10min开始稳定,维持5h以上,但血浆样品和酚试剂产生的颜色在45min以后才能稳定。本方法的回收率为100.28%±1.86%(n=4),变异系数为4.33%用本法和经典的2,4-二硝基苯肼法同时测定16份正常成年人血浆中Vit C含量,结果一致(r=0.98,P<0.01)。与以前所用方法比较,本法具有简便、灵敏(最低检出浓度0.57μmol/L)、快速(60min)、准确等优点。作者还提出了用本法评价人体Vit C营养状况的参考标准。This article represents an experimental research of a colourmetric method for the estimation of vitamin C by using phenol reagent The result shows that absorption maximum of wavelength of the blue compound gained by interaction of ascorbic acid with phenol reagent is 750 nm. There is a positive relation between the optical density and Vit C concentration in the range of concentration of 1.14-45.4 μmol/L The colour produced by vitamin C standard material and the phenol reagent was stable from 10 minutes to 5 hours, but the colour produced by samples of plasma interacting with the phenol reagent became stable 45 minutes later. The recovery rate of the method is 98.2%-102.8%, and the coefficient of variation of precision test is 4.33%. The value of 16 samples of normal human plasma obtained by the method was very closed to the value obtained by classical 2,4-dinitrophenylhydrazin method at the same time (r=0.98, P<0.01) and is in accord with what is reported in the literature.
