摘 要:目的应用HPLC法建立新面醑中甲硝唑和氯霉素含量测定方法。方法采用HPLC法测定新面醑合剂中甲硝唑和氯霉素含量,色谱柱:Extend-C18(250 mm×4.6 mm,5μm),流动相:乙腈-0.05 mol/L磷酸二氢钠溶液(用磷酸调pH至2.5)(55∶45),检测波长274 nm;柱温40℃。结果甲硝唑在54~162μg/ml范围呈良好的线性关系,r=0.9999;氯霉素在48~144μg/ml范围呈良好的线性关系,r=1。结论 HPLC法能准确测定出新面醑合剂中甲硝唑和氯霉素含量,且方法简单、方便、专属性高,适合该制剂的含量测定。Objective To establish a determination method of metronidazole and chloramphenicol contents in Xinmianxu by HPLC.Methods HPLC was used to determine the contents of metronidazole and alficetin in the Xinminxu composition.The chromatographic column was Extend-C18(250 mm × 4.6 mm,5 μm);the mobile phase was acetonitrile-0.05 mol / L sodium dihydrogen phosphate solution(the pH value was adjusted with phosphoric acid to 2.5)(55: 45);the detection wavelength was 274 nm;the column temperature was 40 ℃.Results Within the range of 54-162 μg / ml,there was a good linear relationship of metronidazole(r = 0.9999).Chloramphenicol showed a good linear relationship within the range of 48-144 μg / ml(r = 1).Conclusion The method of HPLC can accurately detect the contents of metronidazole and chloramphenicol in the Xinmianxu composition.The method is so simple and convenient with a high specificity that it is suitable for the content determination.
