摘  要:目的探讨肾上腺素β3受体(β3-AR)在膀胱出口梗阻(BOO)型逼尿肌不稳定(DI)中的作用。方法复制大鼠BOO型逼尿肌不稳定动物模型,6周后行充盈性膀胱测压,根据是否有DI将下尿路梗阻大鼠分为不稳定(DI)组和逼尿肌稳定组,离体逼尿肌条拉力实验观察选择性β3受体激动剂BRL37344A对逼尿肌自律性和舒张功能的影响,尿动力仪记录静脉注射BRI37344A(1 mg/kg及5 mg/kg)对DI大鼠膀胱最大容积及顺应性的影响。结果静脉注射BRL37344A(5 mg/kg)可明显增加正常及逼尿肌不稳定动物膀胱最大容量和顺应性。BRL37344A能明显抑制大鼠膀胱逼尿肌收缩强度,其最大抑制作用分别是对照组(68.5±3.7)%,不稳定组(47.6±5.8)%,稳定组(66.5±7.7)%,对不稳定膀胱组的作用强度明显低于对照组及梗阻后稳定组(P<0.05)。结论β3-AR激动剂可能用于治疗不稳定膀胱。【Objective】 To investigate the role of β3-adrenoceptors(β3-AR) in rat detrusor instability secondary to bladder outflow obstruction(BOO).【Methods】 Animal models of detrusor instability were made in female rats,the effects of β3-AR agonists BRL37344A on the frequency and relaxation response to isolated detrusor were examined by means of the in vitro detrusor strip study.and the effect of BRL37344A on urinary bladder function was investigated by cystometry.【Results】 There was a significant decrease in the density of β3-adrenoceptors in the instability group compared with the controls and the stability group(P 0.05).The intravenous BRL37344A significantly increased bladder capacity.BRL37344A can decrease the contraction amplitade of the isolated detrusor,the potency did differ significantly between normal or stability and instability group.【Conclusions】 β3-adrenoceptors agonists may be effective in the treatment of urinary frequency.
