摘  要:目的研究DNCB诱导小鼠耳肿胀试验中小鼠耳肿胀程度与不同组织内IL-4和IFN-γ表达的相关性。方法以0.50%的DNCB溶液于小鼠背部诱导致敏3d,7d后测量小鼠双耳厚度并分组,以2.00%,1.00%,0.50%,0.25%,0.10%,0.05%和0七种浓度的DNCB溶液于各组小鼠左耳激发,右耳作为对照。每24h测量小鼠双耳厚度并于激发后72h处死小鼠,取血液、双侧耳组织、两侧耳部回流淋巴结,测定组织内IL-4和IFN-γ的表达。结果小鼠激发侧耳肿胀指数与DNCB激发浓度成线性正相关(P<0.01)。小鼠激发侧耳组织内IL-4高表达,并与激发后48h耳肿胀指数正相关,相关系数为0.78(P<0.01)。IFN-γ与激发后48h耳肿胀指数相关系数较低或不相关。结论小鼠耳肿胀程度与DNCB激发浓度、激发侧耳组织内IL-4存在相关性。Objective To detect the expression of IL-4 and INF-γ in different tissue in mouse ear swelling test induced by DNCB. Methods The mice were induced by 0.50% DNCB solutions on the back for 3 days. The mice were grouped and measured both side ears thickness 7 days later, and then the mice were challenged by 2.00%, 1.00%, 0.50%, 0.25%, 0.10% , 0.05% and 0 DNCB solutions on left side ear, it's right side was used as control. Ear thickness was measured every 24h. Mice were killed at 72h late, IL-4 and INF-γ were measured from the sample of blood, ear tissue and local drained lymph nodes. Results The ear swelling and the challenging concentration of DNCB had a positive correlation(P 〈 0.01 ). IL-4 was high expression in the ear tissue of challenged side, and IL-4 in the ear tissue had a positive correlation to the ear swelling of 48h late after challenge(P 〈 0.01 ). However, INF-γ was low expression and the correlation coefficient to the ear swelling was low or uncorrelated. Conclusion The ear swelling is correlated to the challenging concentration of DNCB and the expressions of IL-4 in the ear tissue.
